Background and Objective: Micro RNAs(mi RNAs) are a class of widely distributed, highly conserved non-coding sequences of single-stranded small RNA molecules, the length are 18 to 25 nucleotides(nt), the mature mi RNAs mainly through m RNA base pairing with the target genes to form a silencing complex(RNA-induced silencing complex, RISC) to degradation the target gene m RNAs or hinder their translation, regulate the target gene expression at the post-transcriptional levels, play an important role in embryonic development, cell proliferation, differentiation, apoptosis and tumor development and progression by silencing target genes. It has found that mi R-339-5p was different levels down expressed in breast cancer, colorectal cancer and non-small cell lung cancer and some other malignancies by reading the literatures, it inhibited the development and progression of tumor. So our group select 123 cases of diffuse large B-cell lymphoma, detect the expression of mi R-339-3p and mi R-339-5p in diffuse large B-cell lymphoma and explore its relationship with clinicopathological features of diffuse large B-cell lymphomaMethod :Detect the expression of mi R-339-3p and mi R-339-5p in 123 cases of diffuse large B-cell Lymphoma and 20 cases of reactive lymphoid hyperplasia tissues by using the technology of chromogenic in situ hybridization, and detect the expression of Ki-67 and BCL-6 in DLBCL tissues.Results(1) The positive rates of mi R-339-3p in DLBCL and RH tissues were 34.5%(29/84) and 70%(14/20), the positive rates of mi R-339-5p in DLBCL and RH tissues were39.8%(49/123) and 90%(18/20), the expression of mi R-339-3p and mi R-339-5p in DLBCL tissues were significantly lower than the RH tissues(P <0.01).(2) The expression of mi R-339-3p and mi R-339-5p were significantly reduced in the group of patients that who were lymphoma Ann Arbor stage( Ⅲ ~ Ⅳ) or International Prognostic Index(IPI) score high-risk, but irrespective of sex, extranodal lesions, age, tumor sizes and LDH; the positive of mi R-339-5p in GCB type DLBCL51.9%(27/52) was significantly higher than that in ABC type DLBCL patients 31.0%(22/71)(P <0.05).(3) The difference between the survival rate of positived and negatived of mi R-339-3p in ABC type and GCB type DLBCL patients was no significant statistically(P> 0.05); the survival rate of negatived patients was significantly lower than the survival rate of positived patients in ABC type and GCB type DLBCL patients(P﹤0.01).(4) The positive rate of BCL-6 in mi R-339-5p positived patients 28.6(14/29)was significantly lower than that in mi R-339-5p negatived patients 74.3%(55/74)(P<0.01),the expression of BCL-6 was significantly negatively correlated with the expression of mi R-339-5p(rs =-2.420, P <0.001)。The difference between the positive rate of BCL-6 in mi R-339-5p positived 57.7%(15/26) and negatived patients 63.8%(37/58)was no statistically significantly,the expression of BCL-6 was not significant correlated with the expression of mi R-339-3p(rs=0.054,P>0.05)。Conclusion(1) The low expression of mi R-339-3p and mi R-339-5p might be involved with the occurrence and infiltration progression of DLBCL, the low expression of mi R-339-5p might be related with the poor prognosis.(2) BCL-6 might be involved with the occurrence of DLBCL by playing a role as thedownstream target genes of mi R-339-5p.
