Objective:To Study the pretreatment with prostaglandin E1( PGE1) on acute superior mesenteric artery(SMA) ischemia reperfusion injury of intestinal cell in rats.Methods:18 healthy male SD rats were randomly divided into control group, model group and experimental group. Fasting for 12 hours before the experiment,free drinking water.After anesthesia,From the abdominal median incision into the abdomen.IRI of SMA model was made by clamping the SMA for one hour and two hours after reperfusion in model group and experimental group,respectively.PGE1(20ug/kg) was injected from the tail vein in experimental group.The abdomen was closed control group separation after exposure of SMA,and don’t do anything.In the model building period,To the intraperitoneal injection of 15~20ml/kg Saline.A low blood volume reaction to prevent reperfusion.After abdominal closure.To the intraperitoneal injection of 25~30ml/kg saline(containing 320000 U of gentamicin /L).For expansion and prevent the possibility of infection of abdominal cavity.After two hours of reperfusion specimens and blood.Character of pathology of small intestine was examed. The expression of Bax and Bcl – 2 in small intestine cells and Change of IFABP and DAO in serum were detected.Results:(1).HE detection:pathologic changes showed that there was no change in control group;.while in model group,chorionic epithelium mucosa ulceration and hemorrhage and necrosis occurred than that in experimental group.Experimental group Chiu’sscore was significantly lower than that of model group.(2).Expression of BAX、BCL-2:was higher in model group and experimental group than that in control group(P<0.05), and it was obviously lower in model group than in experimental group(P<0.05).The ratio of BCL-2/BAX:the experimental group was significantly higher than that in model group(P<0.05)(3).The content of IFABP in serum:The content of serum IFABP of three groups before ischemia is basically the same(P>0.05).It was higher in model group and experimental group than control group(P<0.05), and it was lower in model group than in experimental group(P<0.05).(4).The content of DAO in serum:Serum DAO in three groups before ischemia change slightly.It was higher in model group and experimental group than in control group(P<0.05), and it was lower in model group than in experimental group(P<0.05).Conclusions:PGE1 can relieve the alvine necrosis caused by rats’ mesenteric reperfusion injury after acute artery ischemia and thus protect damaged mucosa of small intestine.
