Background: The angioimmunoblastic T-cell lymphoma(AITL) is a unique subtype of peripheral T-cell lymphoma(PTCL), has a unique clinicopathologic features. The study found that virus infection, genetics and autoimmune abnormalities change is closely related to its pathogenesis. There is often accompanied by EBV infection in AITL, EBV may play an important role in the pathogenesis, but the exact mechanism is unknown. Meanwhile EBV is closely related to the progression and prognosis of AITL. PTEN, as one of the most common tumor suppressor gene, are closely related with tumors and hematological tumors. PTEN mutation mainly resulting in the activate of PI3 K / Akt pathway, leading to tumors and autoimmune and other diseases. There are many factors related to the mutations of Pten. EBV latent gene products may affect the expression of host gene, the researchers found that PI3 K / AKT pathway plays an important role in EBV-associated tumors. EBV latent membrane protein LMP1 and LMP2 can activate the PI3 K / AKT pathway, which plays an important role in the development of tumors. Angiogenesis is an important part of tumor growth, invasion and metastasis. The vascular endothelial growth factor(VEGF) is the most important tumor angiogenesis cytokine. One of AITL pathological features is the high expression of VEGF, meanwhile VEGF is adjusted by many factors, such as EBV and PTEN.Aims: The study is to discuss the expression and clinical pathological significance of EB virus(Epstein-Barr virus, EBV), PTEN and VEGF in Angioimmunoblastic T-cell lymphoma(AITL) cancers.Methods: EBV-encoded small RNA(EBER) expression in 21 cases of AITL were detected with in situ hybridization. Detected the expression of 21 cases AITL and 20 cases of lymph node reactive hyperplasia of PTEN and VEGF by immunohistochemical En Vision two-steps method. The expression and clinicopathological significance of EBV, PTEN and VEGF in AITL were analyzed.Results: In 21 cases of AITL, the positive expression rate of EBER was 61.9%; Differential expression of PTEN and VEGF in AITL and lymph node reactive hyperplasia were significant(P<0.05). EBER and PTEN expression was negatively correlated(P <0.05).(2) In Angioimmunoblastic T-cell lymphoma, the EBER positive expression rate of male patients and the progressed group was 80 % and 78.6% respectively, significantly higher than female patients and non- advanced group(P<0.05); the PTEN expression rates in the accompanying B symptoms and progressed group was 31.3% and 21.4%, significantly lower than those without B symptoms and non- advanced group(P <0.05).(3) Survival analysis showed that the PTEN expression was negatively correlated with the overall survival of patients(P <0.05).Conclusion: Whether the EBV involved in the occurring of Angioimmunoblastic T-cell lymphoma by down-regulating PTEN expression is unclear, further research is needed to be explored. EBV infection and low expression of PTEN may indicate the deterioration progress of angioimmunoblastic T-cell lymphoma.
