Objective:To investigate curative effect of remnant tension and augmented suture with semitendinosus tendon autografts in rabbits for the clinical treatment of acute complete rupture of anterior cruciate ligament. Methods:Thirty healthy female adult New Zealand white rabbits, weighting 1.9-2.5kg,averaging2.3kg,Acute complete ACL femoral rupture model of both two knee joints were created in 27 rabbits. The ACL were repaired by remnant tension and augmented suture with semitendinosus tendon autografts in all left knees(L group),All right knees were subjected to anterior cruciate ligament reconstruction (ACLR) with semitendinosus tendon autografts(R group),the rest were selected as normal specimens.Nine animals were sacrificed randomly in deep general anesthesia with an overdose of pentobarbital sodium at 3th,6th,10th postoprative week,The remnant-to-graft and tendon-bone healing healing capacity was depended on macroscopic evaluation and histological examination, and the biomechanical test determined the average maximum load (in Newtons). Results:There were no postoperative complications such as infection and nonunion happened. After three weeks of operation,The peripheral of grafts in the group R had formation of little fibroblasts, and the tendon and bone was connected loosely with inflammatory cells; The fibroblasts of peripheral of grafts in the group L were more than group R,most of the tendon and bone was connected tightly with granulation tissue and a small amount of vascular tissue; After six weeks of operation,The peripheral of grafts in the group R had formation of some fibroblasts, and the tendon and bone was connected tightly with granulation tissue; The central of grafts in the group L had formation of some fibroblasts, the tendon-bone interface appear Sharpey’s fiber formation, and a number of vascular tissue; After ten weeks of operation, The central of grafts in the group R had formation of some fibroblasts, and the tendon and bone was connected tightly, with Sharpey’s fiber appear; The peripheral and central of grafts in the group L had formation of a number of fibroblasts, the tendon-bone interface appear Sharpey’s fiber formation, and some chondrocytes;Biomechanical analysis displayed that the average maximum load of the group R was better than that of the group L(P< 0.05),however,as the increasing observation time, the gap between the both group has gradually narrowed..Conclusion: The ACL reconstruction by remnant tension and augmented suture with semitendinosus tendon autografts in rabbits is more beneficial to histological maturity of the ACL graft than traditional ACL reconstruction, but the tensile strength is not enough strong in the early time.
