Exploring The Action Mechanism Of The Treatment Of “qutanhuoxue Granules”for NAFLD Model Rats Based On The Theory Of Glycerin Water Channel Protein

Objective: The experiment aims to research the influence of body weight, serum biochemical indexes(ALT,AST,GGT,TC,TG), liver pathology,the expression of AQP-7on fat cells and AQP9 on liver cells of nonalcoholic fatty liver(NAFLD) model rats after being treated by“Qutanhuoxue granules” and to explore the possible mechanism of treatment of “Qutanhuoxue granules” in NAFLD.Methods:Choosing 55 healthy male SD rats in 190-250 g,then all the rats are divided into 5 groups randomly(normal group,model control group,low dose group,medium dose group,high dose group)after adaptive breeding for 1 week.Normal control group were fed with common diet,and the other 4 groups were fed with high fat diet.One or two rats were killed randomly in 6th,8th and 10 th week. Confirming whether the model was successful or not by the liver HE staining. After confirming that the model was successful,all the rats except normal group received lavage according to the above group once a day and are killed in day 14,day 21 and day 28. Then observing the pathological changes of liver tissue through the HE staining,determinating liver function(including ALT,AST,GGT) and blood lipid(including TC、TG),detecting the expression of AQP-9m RNA in liver and AQP-7 m RNA in adipose tissue by Realtime PCR.Results:During the experiment,the weight of the normal group is lower than the other groups.After 10 weeks for traditional Chinese medicine treatment,weight growth rate of model group was obviously higher than that of low,medium and high dose group.The difference of the growth rate between low、 medium and high dose group is not obvious(P>0.05). HE staining: Compared with normal group,the liver steatosis of model group were significantly aggravated(P<0.05). Compared with model group,the difference of the liver steatosis between low,medium and high dose group is not obvious(P>0.05).Elisa:Compared with normal group,model group serum AST,ALT,GGT,TC and TG were significantly increased(P<0.05). Compared with the model group,the serum AST,ALT,GGT,TC and TG of low, medium and high dose group were different degree of decline,the difference was statistically significant(P<0.05). But they has not been restored to normal levels,compared with normal group,the difference is statistically significant(P<0.05). Overall: Compared with low dose group,the ALT,GGT of middle and high dose dose group decrease more significantly(P<0.05). And there was no statistically significant difference between the middle dose group and high dose group(P>0.05). Compared with low,medium dose group,AST,TC, TG of high dose decrease more significantly(P<0.05).Compared with 14 d group and 21 d group, the serum AST, ALT, the GGT,TC, TG of 28 d group decrease more significantly(P<0.05).Compared with 14 d treatment group and 21 d treatment group, the serum AST,ALT,GGT,TC,TG of 28 d group decreased more significantly(P<0.05). Realtime-PCR:Compared with normal group, AQP9 m RNA expression of liver in model group increase and AQP-7m RNA expression of fat in model group decrease, the difference was statistically significant(P<0.05). Compared with model group, AQP9 m RNA expression of liver decrease and AQP-7m RNA expression of fat in low,middle and high dose group increase in different degree(P<0.05).Conclusion: 1.“Qutanhuoxue granules” can inhibit the weight growth rate of NAFLD model rats, reduce NAFLD rat fatty degeneration and restore NAFLD rat pathologic structure in different degree. 2.“Qutanhuoxue granules” can improve liver result enzymology indexes of NAFLD model rat(including ALT, AST, GGT) and also can significantly reduce serum TG, TC level of NAFLD rat model.The treatment curative effect of the high dose group and 28 d treatment group is best.3.“Qutanhuoxue granules” can reduce AQP9 m RNA expression level of liver and elevated AQP-7m RNA expression level of adipose tissue in NAFLD model rat, and restore normal liver lipid metabolic balance, thus reducing the accumulation of glycerol in the model of rat liver. Inhibiting the expression of AQP9and promoting expression of AQP-7may be one of the action mechanism of the treatment of “Qutanhuoxue granules”for NAFLD.
