Objective:This study detected the expression of CA in the molecular Ⅸsubtype of breast cancer and discussed it’s significance and relationship between three drug-resistant protein: Pgp, GSTπ, Topo II-α and p53, so that it could provide more theoretical basis for the clinical application of CAⅨ in breast cancer and its molecular subtype.Methods:The e x p r e s s i o n o f E R 、 P R 、 H e r- 2 a n d K i- 6 7 w a s d e t e c t e d b y immunohistochemical method in pathological spaceman of breast cancer patients, according to the St.Gallen classification standard,breast cancer were divided into four molecular subtype:luminal A subtype、luminal B s u b t y p e、 H e r- 2 o v e r e x p r e s s i o n s u b ty e a n d t r i p l e- n e g a t i v e subtype.Randomly selected 100 cases of every different molecular subtypes, a total of 400 cases,Some clinical and pathological data was s e l e c t e d i n c l u d i n g a g e, B M I, m e n s t r u a l h i s t o r y, r e p r o d u c t i v e history, history of exposure to radioactive substances,breast and gynecological disease history, the history of long-term intake of estrogen a n d p r o g e s t e r o n e, t h e s i z e o f p r i m a r y t u m o r, l y m p h n o d e metastasis,WHO histological grade, and then analysis the clinical andpathological characteristics of different molecular subtypes type,The SP method of immunohistochemistry(streptavidin peroxidase connection method of biological antibiotic) was used to detect the expression of CAⅨ、Pgp、GST-π、Topo II-α and p53 in every case,Then the study explored the expression of CAIX and Pgp, GSTπ, Topo II-αand the relationship between CAⅨ and three drug-resistant markers as well as p53 and Ki-67 in different molecular subtypes of breast cancer.Results:1.400 breast cancer patients’ clinical and pathological data was collected.Analysis the relevant clinical and pathological features parameters of different molecular subtypes of breast cancer species,there is a statistical significance in lymph node metastasis and WHO histological grade,(P<0.05),but not in primary tumor size(P>0.05).2.The expression of CAIX was detected in different molecular subype of breast cancer tissues,the expression of CAⅨ was divided into 0,+,++,+++ groups.it was statistically significantin in different molecular sbbtypes( P< 0.05).The CAIX expression had no statistical significant relationship with patients’ age,lymphnode metastasis,tumor grading,but in size 2cm-3cm tumor,it had a negative correlation,3.Among luminal A, luminal B, Her-2 over expression subtype and TNBC,Pgp resistant patients ratio were respectively 14%,8%,24%,30%(P<0.05),the ratio of Topo II-α drug-resistance were: 88%,54%,44%,26%(P<0.05), the ratio of GSTπ resistancewere respectively: 10%,16%,26%,38%(P<0.05), the difference of three drug-resistant index in different molecular subtypes was statistically significant.4.There is no significant relationship between CAⅨ expression level and Pgp, GSTπ,Topo II-α among four molecular subtypes(P > 0.05).5. In luminal A, luminal B, Her-2 over expression type, TNBC subtype, the p53 positive expression rates were: 16%,16%,44%,40%, the difference was statistically significant(P<0.05).In TNBC subtype, the expression of CAⅨ and p53 was significantly correlated(P<0.05).6.In the 0,+,++,+++ group of CAⅨ, it was31.01±5.77%, 24.05±19.60%, 29.17±19.13%, 41.16±23.96%, the difference was statistically significant(P<0.05), the expression rate of Ki-67 was significantly higher in +++ group than the other three groups.7.Drug resistance proportion of Pgp,Topo II-α, GSTπ were:19%,53%, 9.5%,the difference was statistically significant(P<0.05), Topo II-α resistance proportion was significantly higher than that of Pgp and GSTπ.Conclusion:1.The clinical and pathological characteristics is different in molecular subtypes,the luminal types tends to higher differentiated histological and lower lymph node metastasis rate,the Her-2 over expression and TNBC tend to have lower degree of differentiation.2.The expression of CAⅨ is different in molecular subtypes, the TNBC tend to have a higher expression level.In size 2cm-3cm tumor,CAⅨ had a negative correlation with thetumor size.3.The expression of Pgp, GST, Topo II-α and p53 in different molecular subtypes have a differences.4There is no significant relationship between CAIX and Pgp、TopoⅡ-α、GSTπ.5In the expression of drug-resistance protein, Topo II-α resistant proportion were more than Pgp and GSTπ.6.Among every CAⅨ expression group, the +++ group had a higher expression level of Ki-67.7.In the TNBC subtype,expression of p53 and CAⅨ may be positively correlated, CAⅨ as a hypoxia and acidification index of tumor microenvironment, the detection is simple and might be used in tumor molecular classification.
