Objective Dermal collagen fibers formation is a ordered process by the stepwise assembly of collagen.Collagen monomers formed a stepped longitudinal displacement ordered collagen fibrils. If abnormal collagen arrangement, the assembly and interconnection,the formation of abnormal collagen scaffold structure,the performance for scarring after wound healing.Rabbit ear integration model is established,around the wound respectively were injected with normal saline,decorin(DCN) and decorin antibody (DCN antibody) in the wound healing process.1.Comparing regenerative healing and scar healing morphological differences after wound healing.2.To observe the characteristics of type â…  and type â…¢ collagen in the shallow and deep side.3.To observe the constructive relationships between type â…  and type â…¢ collagen.4.Object to the impact of DCN to the constructive relationships between type â…  and type â…¢ collagen.5.Analysis of collagen fiber in the shallow and deep side different change rules in the process of wound healing, and the impact of DCN to its structure.To provide some more morphological basis to further studies on skin regenerative healing and scar healing aspects.Materials and methods 1.Fifty Japanese white rabbits,With same gender,ageand weight of two white rabbit into a pair of,so basically the same white rabbit can match up to 25,then each pair were assigned to different in the two groups,respectively DCN and DCN antibody group,each group of 25.On only one side of each group of rabbit ears injection of physiological saline as control,therefore see aside of injection of saline as control group.In the ear abdominal do 1cm* 1cm of the wound,from shallow to deep direction were randomly selected from tip of ear to ear or from ear to ear tip do oblique section.The deepest to cartilage layer and cut the perichondrium,so establishment of regeneration and scar of rabbit ear skin wound healing integration model.At 0th,4th,8th,12th and 30th day after wounded,were observed and recorded the wound healing and scar formation, and archive photographs.Group DCN:immediately after injury, each rabbit left ear with 30G needle injection of DCN,and the concentration of DCN was 5g/ml, and each wound dosage is 0.1ml,once every other day,until the wounds healed. The right ear all the wounds with 30G needle injection physiological saline as control.DCN antibody group: immediately after injury,each rabbit left ear with 30G needle injection of DCN antibody,and the concentration of DCN antibody was 10g/ml,and each wound dosage is 0.1ml,once every other day, until the wounds healed.The right ear all the wounds with 30G needle injection physiological saline as control.All the wounds were bandaged with sterile excipients.Compared the average healing time of the wound and observe the morphological changes.2.Application of HE,IHC and computer image processing system,the detection of Japanese big ear rabbit ear ventral skin wound model of external physiological saline and DCN and DCN antibody in before and afte, the expression of type I and type III collagen in different shades of healing wounds and its constructing relationship.3.By using scanning electron microscopy and computer image processing system,the observation of the variation of collagen fibers in different depth layers,and the effect of DCN on the structure.Results 1.The shallow lateral wound (to the shallow side edge distance less than or equal to 4mm)eighth days after operation,the wound scab skin has been out most,shallow lateral wound has been born again.Thirtieth days after operation,no obvious color difference between shallow lateral and normal skin,and no clear limit, for regenerative healing.But the deep side wound (to the deep side edge distance less than or equal to 4mm) until twelfth days after operation,wound still see some scabby exist,not entirely epithelium.Thirtieth days after operation,wound central hill sample bulge, significantly higher than the leather hard, colour and lustre is deep and surrounding tissue for scar healing.2.1n addition, with same side, injection of DCN solution ear healing faster than was injected with physiological saline ones, the wound healing was (41.2±7.6)% at 4th day, (79.5±6.5)% at 8th day, (93.6±4.2)% at 12th day when used DCN, higher respictivly (36.6±4.3)%, (60.5±3.9)% å'Œ (85.2±4.3)% in the NS group (P<0.05).The wound healing was (22.8±4.5)% at 4th day, (47.5±5.3)% at 8th day, (73.9±2.6)% at 12th day when used DCN antibody,lower respictivly the NS group (P<0.05),and the color more close to normal tissue, scar tissue softer and thinner, and wound healing of injection of DCN antibody solution deep color,the scar thick and hard, significantly higher than that of normal skin tissues.The area of regeneration and scarring of the wound,DCNgroupaverage regeneration area (0.86±0.13)cm2 increasedsignificantly compared with NS group average regeneration area (0.71 ±0.11) cm2 (P<0.05),DCN group average scar area(0.14±0.07)cm2 were significantly lowered compared with NS group average scar area (0.29±0.09) cm2 (P<0.05),DCN antibody group average regeneration area(0.46±0.08) cm2,and average scar area (0.54±0.10) cm2 clear difference with NS group (P<0.05)3.The expression of type â…  and type â…¢ collagen: at each time point deep side wound was higher than that the shallow side (P<0.05); The NS group, DCN group and DCN antibody group has the difference with the expression of type â…  and type â…¢ collagen in side wound depth (P<0.05); The expression of collagen type III between groups there is no difference (P>0.05); The relative value of collagen type â…¢ and collagen type â…  of the wound on rabbits ears, DCN group is the highest, and DCN antibody group is the lowest, difference is obvious (P<0.05)4.Scanning electron microscopy showed that the significant differences between the collagen fibers arranged (P<0.05);Conclusions 1.This experiment has successfully established regeneration-scaring wound healing integration model in rabbit ear skin, the shallow lateral wound realize regenerative healing, and the deep side achieve scar healing.2.Decorin injection plays a regulation â…¢/â…  collagen ratio effect, at the same time, the regulation of collagen fibers assembly, played the prevention of scar formation and to realize the regeneration effect.
