Objective To detect the expression of P-gp and HIF-la in oral squamous cell carcinoma tissue and their relationship, and offer a reference for clinical assessment of the multiple drug resistance during chemotherapy in the oral squamous carcinoma.Methods 1% The expression of P-gp(mdr1) gene and HIF-1 a gene in oral quamous cell carcinoma tissue are detected by RT-PCR.2、The expression of P-gp and HIF-la in oral squamous cell carcinoma tissue are detected by immunohistochenmical technique SP.Results 1、In this experiment,28 specimens of oral squamous cell carcinoma tissue are detected, and 15 specimens can reach the experiment requirement. Under the electrophoresis, the result shows that P-gp(mdrl) gene displayed in 15 specimens while HIF-la gene displayed in 13 specimens, and they co-expression in 13 specimens. Through using chi-square statistic of exact propability in contingency table analysis, there are differences (P<0.05) between P-gp(mdrl) gene and HIF-la gene. The expression of them have a direct ratios(r=0.867).2、In 28 specimens of oral squamous cell carcinoma tissues, HIF-1α showed positive-expression (75%) in 21 specimens and P-gp showed positive-expression in 20 specimens(71.4%). Through chi-square statistic there are no significant differences (P>0.05) in the expression of P-gp、HIF-1α、 sex、tumor TNM stage、pathological gradingand lymph node metastasis. The expression of HIF-1α showed difference in the tumor size (P=0.029) and age (P=0.041). There is a statistically difference between the expression of P-gp and tumor size (P=0.007<0.05) significantly. There are 18 out of 28 specimens of oral squamous cell carcinoma tissues with coherence and consistency of positive expressing rate.Through using chi-square statistic of exact propability in contingency table analysis, there is a significant difference(P<0.05) between P-gp and HIF-1α. The expression of P-gp and HIF-1α showed direct ratios (r=0.548)Conclusions The level of P-gp and HIF-1α expression is highly increasing in oral squamous cell carcinoma tissue simultaneously which have significant correlation, that maybe synergistically mediat the primary drug resistance in oral squamous cell carcinoma. Maybe the mechanism come true is through HIF-1α up-regulated the expression of P-gp, detect the expression of P-gp and HIF-1αmay offer a reference for clinical assessment of the multiple drug resistance during chemotherapy in oral squamous cell carcinoma.
