ObjectiveHyperuricemia (HUA) was caused by uric acid excretion dysfunction and (or) purine metabolic disorder that lead to an elevated blood uric acid in the body. Medicine which comes from natural plant has little side effects in treatment of HUA, suitable for long-term use. Various herbal medicines which can reduce blood uric acid level were reported presently. The composition of YiSuan Compound Tea is familiar with Chinese. They are Hawthorn leaves, Radix Astragali and Sophora flowers and so on. YiSuan Compound Tea can retain more active factor without boiling. This study, based in an ideal hyperuricemia animal model with renal injury, intends to observe the effect of YiSuan Compound Tea in lowering blood uric acid and protecting vascular endothelial cells and kidney. Rare article was reported the treatmen effect of combining traditional Chinese medicine and conventional chemical medicine, this study will also explore the interaction between YiSuan Compound Tea and Allopurinol, in order to provide a scientific basis.MethodsFeeding adenine in 25 mg/Kg and oxonic aid potassium salt in 300 mg/Kg per day to establish a hyperuricemia animal model.84 male SD rats were randomly divided into normal control group, model group, Allopurinol group, high dose of tea group, low dose of tea, Allopurinol+high dose of tea group, Allopurinol+low dose of tea group. Except normal control group, each group were feeded the agent for three weeks to establish a hyperuricemia animal model.At the same time, Allopurinol group, high dose of tea group, low dose of tea group, Allopurinol+high dose of tea group, Allopurinol+low dose of tea group were gave the corresponding drugs. We observed the changes of uric acid (UA), nitric oxide (NO), blood urea nitrogen (BUN), creatinine (Cr), cystatin C (CysC), and analyzed the interactions between the two drugs.Results1.The levels of UA, BUN, Cr, CysC were decreased significantly in high dose of tea group, while the NO were higher(P< 0.05).2.The levels of UA, BUN were lower in low dose of tea group, while the NO were significantly higher(P< 0.05).3.The levels of UA, BUN, Cr were decreased significantly in Allopurinol+high dose of tea group and Allopurinol+low dose of tea group, while the NO were higher(P< 0.05). Drug combination can make a interaction in BUN. Low concentration of YiSuan Compound Tea and Allopurinol share to reduce BUN is more effectively than used them separately.Conclusion1. YiSuan Compound Tea can have the same effect as Allopurinol in reducing uric acid, and protect the vascular endothelial cells. Meanwhile, the high concentrations of YiSuan Compound Tea to improve renal injury is obvious.2. Combining YiSuan Compound Tea and Allopurinol have certain effect for UA, BUN, Cr, NO, but the synergy is not obvious. Low concentration of YiSuan Compound Tea and Allopurinol share to reduce BUN is more effectively than used them separately.
