Objective To provide new experimental evidence for the movement to improve the molecular mechanisms of diabetes by investigating the changes of TLR4 and NF-kB and the change of the liver tissue after different intensity exercise on type 2 diabetic rats.Methods (1) Fifty SD rats were divided into control group(NC),type 2 diabetic group(DM),low intensity exercise group(LIE), middle intensity exercise group (MIE)and high intensity exercise group(HIE) randomly,with 10 rats in each group. The rats of NC group were given normal diet,the other four groups were given the diet of high fat and high sugar(regular feed and 5%of egg yolks,10% sucrose and 10% of refined lard).After four weeks,the rats of NC group were injected citric acid buffer.The rats of other four groups were injected low-dose streptozotocin intraperitoneally to establish the model of type 2 diabetes. (2) Tow rats were selected randomly.The longest time of the SD rats can insist on swimming was the exhaustion time.The standard of the longest time is that rats can keep on the surface of the time less than 8 seconds finally.The three exercising groups were trained by swimming in the exhaustion time of 40%、70% and 100%,once a day,6d/w,continuous training for 6 weeks.After the experiment, rats was put to death and blood and liver tissue was selected.Then the levels of factors TLR4 and NF-kB were measured.The level of expression of mRNA of TLR4 andNF-kB of liver were tested by the method of semi-quantitative RT-PCR.The level of expression of protein of TLR4 and NF-kB of liver were tested by the method of immunohistochemistry.Liver tissue was fixed with formalin,embedding with paraffin,colouring with HE dyestuff.Then the pathology morphological changes was analysised by microscopic.Result (1) The liver tissue of rats in each group showed:the liver tissue of NC group didn’t have any abnormality.The liver tissue of the remaining four groups were found pathological changes of the typical diabetes.Compared to the DM group,the liver tissue of rats of the three exercise groups restored in different degrees,and the recovery in the MIE group was most obviously. (2) Compared to NC group,the body weight and the liver weight of the other four groups decreased markedly(P<0.05),the FBG increased obviously(P<0.05),the level of expression of mRNA of TLR4 and NF-kB increased obviously(P<0.05),the level of expression of protein of TLR4 and NF-kB increased obviously (P<0.05);Compared to DM group,the FBG decreased in the there exercise group(P<0.05),the level of expression of mRNA of TLR4 and NF-kB decreased obviously(P<0.05),the level of expression of protein of TLR4 and NF-kB decreased obviously(P<0.05);Compared to LIE group and HIE group,the FBG decreased obviously in MIE group(P<0.05),the level of expression of mRNA of TLR4 and NF-kB decreased obviously(P<0.05),the level of expression of protein of TLR4 and NF-kB decreased obviously(P<0.05).Conclusion 6 weeks of swimming training can reduce the FBG and the level of expression of mRNA and protein of TLR4 and NF-kB of liver of the type 2 diabetic rats,improving the state of inflammation of the liver,recovering the morphology of the liver tissue,slowing down the development of the diabetes of type 2. And the effect of middle-intensity exercise is best.
