Objective To investigate the effect of trimetazidine pretreatment on myocardial injury induced by propofol in rats.Methods Forty healthy SD rats,weighing 240-250 g,aged 2.5 month,were randomly divided into 4 groups (n=10):lipid group (group L),10% lipid was infused for 12 h; propofol group (group P),1% propofol was infused for12 h;group TL and group TH,2 mg·kg-1 or 5mg·kg-1 of trimetazidine were injected intravenously 1.5 h before propofol infusion respectively. Propofol or lipid was infused at a rate of 30 mg·kg-1·h"1 for 12 h. Internal jugular vein catheterization.1.5 ml blood from jugular vein was taken at 6 h and 12 h of propofol infusion. Creatine kinase (CK), creatine kinase (CK-MB), lactate dehydrogenase (LDH), a-hydroxybutyric acid dehydrogenase (a-HBDH),triglycerides (TG), low-density lipoprotein (HDL) and high-density lipoprotein (LDL) were detected using enzyme coupling method, troponin (cTnI) were detected using chemiluminescence enzyme immunoassay. Myocardial tissue was obtained at the end of infusion for pathological examination with both optical and electron microscopy.Results Compared with group L,the serum CK, CK-MB, LDH, a-HBDH, cTnI and TG levels were significantly increased,HDL was decreased in group P and TL (P<0.01),and LDL was no statistical significance(P>0.05),the serum TG, HDL, LDL were increased in group TH (P<0.01), and there was no statistically significant at the rest of the indicators (P>0.05). Compared with group P, the serum CK, CK-MB, LDH, a-HBDH,and cTnI levels were increased in group TL and TH (P<0.01),the serum TG, HDL and LDL were increased in group TH and TG was decreased in group TL (P<0.01). Compared with group TL, the serum CK, CK-MB, LDH, a-HBDH and cTnI were decreased,the serum TG, HDL and LDL were increased in group TH (P<0.01). The main damage observed by electron microscope presented at mitochondria, muscle fibers and Z line, no damage observed in group L.Conclusion Trimetazidine pretreatment can attenuate myocardial injury, but it may increase Lipid level.
