Objective:To investigate the influence of trimetazidine in ovarian cancer mice that received adriamycin (ADM) chemotherapy, by analyzing the change rule of serum cardiac troponin I(cTnI) level and the pathological changes of myocardium observed with microscope and transmission electron microscope, and explore the mechanism of myocardial protection after ADM chemotherapy with TMZ treatment, and to provide some experimental evidence in myocardial protection for the cancer-chemo patients.Methods:The nude mouse models of ovarian cancer were established with innoculation of ovarian cancer cell.32 nude mice were randomly divided into 4 groups:Group A (normal sodium group), group B (ADM+ normal sodium group), group C (ADM+TMZ 20mg/kg/d group), group D (ADM+TMZ 40mg/kg/d group). Group A were administered intraperitoneally with 10ml/kg of normal sodium and group B,C,D were administered intraperitoneally with 2.5mg/kg of ADM, twice a week over a period of 4 weeks. From the third week, the nude mice in group A and B were treated with saline by intragastric administration with equal volume with TMZ in group C and D, once a day over a period of 2 weeks, the nude mice in group C and D were treated with TMZ by intragastric administration 20mg/kg/d,40mg/kg/d respectively. All nude mice were treated with anesthesia by 10% chloraldurat on the second day after the last administration. Blood samples were taken from following respectively:before intragastric administration, before anesthesia and 30 min after anesthesia.The samples were certrifuged after they were put aside 30 min, and the blood serum were kept in -80℃refrigerator. Serum cTnI level were detected with enzyme linked immunosorbent assay. The pathological changes of myocardium were observed through microscope and transmission electron microscope.Results:(1)Serum cTnI level in four groups were statistically significant (P<0.05) at T1. At T2 and T3,cTnI in group C and D were lower than that in group B(P<0.05), but there were no statistically significant differences between group C and D(P>0.05).(2) Serum cTnI level in three points was no statistically significant in group A(P>0.05). In group B, the serum cTnI level at T3 was higher than that at T1 and T2 markedly(P<0.05), but there were no statistically significant differences at T1 ahd T2; The cTnI of each two points were statistically significant in group C and D as follows:T1 to T2, T2 to T3 and T3 to T1(P<0.05).(3)The pathological changes and myocardium ultramicro-structure in group B were extremely serious compared to that in group A. The changes in group C and D were more lighter than that in group B. Conclusion:(1) After adriamycin-chemotherapy, the level of serum cTnI elevated in ovarian cancer nude mice; (2)The level of serum cTnI and the changes of myocardium were decreased with TMZ treatment, which indicated that TMZ has the effect of myocardial preservation.
