Objective:Hyperlipemia is one of the risk factors of AS(athrrosclerosis), and it keeps a close relationship with cardiovascular and cerebrovascular events). a large number of studies show that long-term high blood fat can lead to intimal lesions and inflammatory responses, cause endothelial dysfunction, and act as the common pathological basis of vascular lesions throughout the diseases development). In this study, model of high fat diet feeding guinea pig was used to investigate the effects of compound Chinese medicine Qing Zhi Capsule on the level of four blood lipids and the protein expression of aortic inflammation factors, and explored the mechanism of the relationship between hyperlipemia and cardiovascular and cerebrovascular diseases。Methods: 60 male Dunkin-Hartley guinea pigs were randomly divided into 6 groups, 10 in each group. The normal group was fed with normal maintenance food, the other groups were fed with high lipid food, all the animals took food freely,2 weeks later, the positive control group was given Lipitor(Atorvastatin Calcium Tablets),groups of Fufang Qingzhi capsule low, middle, high dose group(0.2, 0.4, 0.8 g/kg.d-1)were given Fufang Qingzhi respectively for 4 weeks, recorded the body weight 1 time a week. Then the animals were fasted for 10 hours, 10% chloral hydrate was used to induce anesthesia, collected blood through abdominal aortic, separated serum(3000 rpm, 10 minutes); a part of horacic aortas were preserved in liquid nitrogen, another part of the horacic aortas were kept in neutral formalin. Automatic biochemical analyzer was utilized to detect the serum lipid levels and liver function indicators: such as total cholesterol(CHO), serum triglyceride(TG), high density lipoprotein(HDL), low density lipoprotein(LDL), alanine aminotransferase(ALT), aspartate aminotransferase(AST). HE staining of the horacic aortas tissue was performed to evaluate the pathological changes. The protein expressions of the IL-6, IL-10, IL-1beta, TNF-alpha, adhesion molecule-1(ICAM-1) of guinea pig thoracic aorta tissue were detected by Western Blot.Results:1 the general conditions and body weight changesCompared with the normal control group, the general conditions of the animals in high fat diet groups were good, a small number of animals showed decreased appetite, body weight grew slowly. Compared with normal group, the high fat diet groups body weight increased to some extend, but didn’t show significant differences between groups(P>0.05).2 changes in serum four blood lipids and liver functionCompared with the normal group, the total cholesterol(CHO), serum triglyceride(TG), low density lipoprotein(LDL), alanine aminotransferase(ALT), aspartate aminotransferase(AST) in model control group increased significantly(P<0.05), high density lipoprotein(HDL) decreased to some extent, but didn’t show any statistical difference(P>0.05).Compared with the model group, total cholesterol(CHO), serum triglyceride(TG), low density lipoprotein(LDL) in positive control group and Fufang Qingzhi capsule groups were significantly decreased(P< 0.05), alanine aminotransferase(ALT), aspartate aminotransferase(AST) and high density lipoprotein(HDL) were also improved in some degree, but didn’t show any significant difference(P>0.05).3 the observed results of aorta histopathologyNormal control group: the aortic wall showed clear hierarchy, endomembrane was smooth and successive, medial smooth musclelayer was uniform, no shrink, and the thickness is normal, internal elastic membrane and media membrane elastic fiber were clear, the outer layer was loose connective tissue. Model control group: endotheliocyte swelling and hyperplasia, of smooth muscle in tunica intima showed hyperplasia and swelling, middle layer cells arranged in disorder, the lines were not clear. The positive control group: endotheliocyte swelling was visible, vascular wall level was clear, media membrane swelling was visible. Fufang Qingzhi capsule high dose group: endothelial cells were slightly swelling, vascular wall showed clear hierarchy, internal elastic membrane and membrane elastic fibers were obviously visible. Fufang Qingzhi capsule middle dose group: endothelial cells were slightly swelling and accompanied by the increased quantity, the level of the vascular was clear, tunica media slight swelling was visible. Fufang Qingzhi capsule low dose group: the number of endothelial cells increased obviously, arranged intensively, endomembrane was successive on the whole.4 the protein expressions of inflammatory factors in the arterial vascular tissueCompared with the normal group, the protein expression of IL-6, IL-1 beta,TNF- alpha, adhesion molecule-1(ICAM-1) of the model control group were increased significantly(P<0.05), the expression of IL-10 was decreased significantly(P<0.05), the results showed that the hyperlipidemia animal model was built successfully, and satisfied the demand of the experiment.Compared with the model group, positive control group(Li Putuo) and compound Qingzhicapsule could significantly decrease IL-6,IL-1β,TNF- α, adhesion molecule-1(ICAM-1) protein expression, increase the expression of IL-10(P<0.05).Conclusions:1 high fat diet can induce the guinea pig hyperlipemia, and cause inflammation injury and dysfunction in vascular endothelial cells, the model can be used for relevant experimental study.2 compound Qingzhi capsule can regulate lipid metabolism, improve endothelial function, inhibit inflammatory reaction, showed protective effect on hyperlipidemia guinea pig aorta.
