Compatibility of traditional Chinese medicines, the main method of clinical medical therapy, can help to create new treatment effects, evaluate pharmacodynamics effects, reduce the toxicity of traditional Chinese medicines, and function in multiple symptoms by combining different medicine together accordingly. The combination of Cassia Twig and Radix Paeoniae Alba, which was widely used by Zhongjing Zhang, also turned up to the frequency of 27 times in his famous masterpiece〈Treatise on Febrile and Miscellaneous Disease〉. Cassia Twig, with the property of Xin, Gan and Wen can be used to treat disease caused by cold factors. Xin and Wen can keep Ying and Weiqi in balance and release the body. Gan and Wen can help strengthen Yang and change Qi. Enough Yang ensures sufficient energy and vitality. The lienogastric system works well; the cold factor can be dispelled.Radix Paeoniae Alba, with the property of Ku, Suan and Han, mainly works on liver channel, can be used to nourish blood, astringe Yin and dredge the stasis, can be effectively used to abatement fever, relieving restlessness and relieve spasm and pain.Cassia Twig can work on reduce symptoms and Radix Paeoniae Alba can help to consolidate patients’ body. Cassia Twig can help to create vitality and Radix Paeoniae Alba can help to relieving restlessness. Cassia Twig can treat the exterior syndromes and Radix Paeoniae Alba works on interior syndromes. Cassia Twig and Radix Paeoniae Alba can act at each other’s properties and create medical effect while combined in right dosages. The combination can increase diaphoretic function meanwhile arrest sweating properly. Also the combination can reduce patient’s symptoms and improve patient’s physical condition. Over all, the combination of Cassia Twig and Radix Paeoniae Alba approve obvious effect on balancing and releasing the body, increasing diaphoretic function and relieving spasm and pain.“The key secret which never been released about traditional Chinese medicine is the right dosage”. The effect of medicine is measured by dosage. The effect of traditional Chinese medicine depends on the right dosage. Although Zhongjing Zhang used the combination of Cassia Twig and Radix Paeoniae Alba frequently, he changed the ratio of dosages according to patients’ specific symptoms. To fully understand the effects of combination of Cassia Twig and Radix Paeoniae Alba on diaphoretic function and pain-relieving function in Zhongjing Zhang’s different formulas, the dosage ratio of these two herbs is worthy researching. This essay will research the influence of dosage ratio of the combination of Cassia Twig and Radix Paeoniae Alba effect on diaphoretic function by observing the quantity of plantar sweat gland spots, the amount of sweat and the incidence rate of sweat gland vacuole of SD rats. To observe the influence of different dosages of Cassia Twig on rats’ intestinal propulsion, rats which have diarrhea under the influence of Dachengqi Decoction will be the experimental objects. At the same time, the extraction ratio of Paeoniflorin produced by different dosage ratios of combination of Cassia Twig and Radix Paeoniae Alba and the effect of Paeoniflorin on pain-relieving function will be researched in order to get a better understanding of the combination of Cassia Twig and Radix Paeoniae Alba and provides necessary help for clinical treatment.Part one Experimental study on different dosages of combination of Cassia Twig and Radix Paeoniae Alba on diaphoretic functionObjective: To observe the different dosages of combination of Cassia Twig and Radix Paeoniae Alba on rats’ diaphoretic function.Methods:(1) Divide 120 SD rats into 6 groups equally and randomly. Group A was blank. Group B was Cassia Twig. Group C was Radix Paeoniae Alba. Group D, the combination of Cassia Twig and Radix Paeoniae, was specifically divided into Group D1, combine Cassia Twig and Radix Paeoniae Alba in ratio of 1:0.5, Group D2, combine Cassia Twig and Radix Paeoniae Alba in ratio of 1:1 and Group D3, combine Cassia Twig and Radix Paeoniae Alba in ratio of 1:2. Provide the same experimental environment to all the groups: the temperature of the lab is(25±1) ℃and the temperature of medicine is(35℃±2℃). Observe the quantity of plantar sweat gland spots, the amount of sweat of the SD rats in every group after intaking different medicine accordingly and separately. Observe the plantar sweat gland and record the number of sweat gland cells and physalides to count the incidence rate of sweat gland vacuole after soaking the plantar foot pads, skin and subcutaneous tissue into 4% Paraformaldehyde solution, paraffine embedding and HE staining 30 minutes after the rats intaking medicine accordingly.(2)Using the High Performance Liquid Chromatography(HPLC) to research the decocting quantity change of Paeoniflorin on different combination of Cassia Twig and Radix Paeoniae Alba.Result:(1)Compare the quantity of plantar sweat gland spots, the amount of sweat and the incidence rate of sweat gland vacuole of the SD rats: Under the premise of the same solution and environment temperature, compared Group A, Group B and Group D showed more plantar sweat gland spot, higher amount of sweat no matter the qualitative or quantitative experiment,and the incidence rate of sweat gland vacuole.(P<0.05). The diaphoretic function was obviously increased in Group B when compared with Group D(P<0.05). The diaphoretic function of group D1 is better than group D2 and D3(P<0.05),group D2 better than group D3(P<0.05).Group A showed higher amount of sweat and the incidence rate of sweat gland vacuole than Group C meanwhile the two groups showed equal plantar sweat gland spots.(2)Compare the incidence rate of sweat gland vacuole: Few sweat gland vacuoles can be observed in Group A and sweat gland diameter were small. Compared with Group A, the sweat gland numbers of vacuole were increased and the sweat gland diameter was obviously expanded in Group B and Group D, and compared with Group D, Group B showed higher incidence rate of sweat gland vacuole and sweat gland diameter. Group D1 showed higher incidence rate of sweat gland vacuole and weat gland diameter compared with Group D2 and D3. compared with Group D3, Group D2 showed higher incidence rate of sweat gland vacuole and sweat gland diameter.Compared with Group A, there was no obvious change of sweat gland vacuole and sweat gland diameter was small in Group C.Conclusion:(1)The diaphoretic function of the Cassia Twig group was significantly enhanced no matter the qualitative or quantitative experiment. The groups which were given Cassia Twig showed more plantar sweat gland spots, higher amount of sweat and the higher incidence rate of sweat gland vacuole.(2)The diaphoretic function can be more obviously observed in the groups which were given Cassia Twig than which were given Radix Paeoniae Alba.(3)Compared among the groups which were given the combination Cassia Twig and Radix Paeoniae Alba, the diaphoretic function was increased when Cassia Twig and Radix Paeoniae Alba were combined in ratio of 1:0.5.(4)When compared with the blank group, the Radix Paeoniae Alba group approved the function of arresting sweat.(5)The experiment results showed that the combination of Cassia Twig and Radix Paeoniae Alba can increased the diaphoretic function. There is a positive correlation between the diaphoretic function and the dosage of Cassia Twig in the combination. And there is a negative correlation between the diaphoretic function and the dosage of Radix Paeoniae Alba in the combination. Among all the groups, the Cassia Twig group showed the most obvious and significant effect on increasing diaphoretic function.Part Two Experimental study on different dosages of combination of Cassia Twig and Radix Paeoniae Alba on pain-relieving functionObjective: To observe the influence of combination of Cassia Twig and Radix Paeoniae Alba on movement of carbon powder in rats’ intestines. To research the decocting quantity change of Paeoniflorin on different combination of Cassia Twig and Radix Paeoniae Alba.Methods: Randomly divide 120 SD rats equally into A(blank), B(Cassia Twig), C(Radix Paeoniae Alba) and D(combination of Cassia Twig and Radix Paeoniae Alba, which was divided into D1(Combine Cassia Twig and Radix Paeoniae Alba in ratio 1:0.5), D2(Combine Cassia Twig and Radix Paeoniae Alba in ratio 1:1), D3(Combine Cassia Twig and Radix Paeoniae Alba in ratio 1:2). Keep the SD rats in the same experimental environment, keep the temperature of the lab is(25±1) and the temperature of medicine is ℃(35℃±2℃). When the experiment was begun, all the rats were given dachengqi decoction(2.7g/Kg),10 minutes after, different groups were given intragastric administration and then different dosages of medicine combinations accordingly. Group A was given normal saline. Then 20 minutes later, all were given carbon powder intragastric administration. At last, another 20 minutes later, the effect on decreasing the rats’ intestinal propulsion can be observed and compared. To research the decocting quantity change of Paeoniflorin on different combination of Cassia Twig and Radix Paeoniae Alba by using the High Performance Liquid Chromatography(HPLC).Results:(1)Under the premise of the same solution and environment temperature, Compared with Group A, Group B, Group C and Group D all approved the decrease function on the carbon powder propulsion of rat intestine in different degrees(P < 0.05); Compared Group B, Group D1 approved obvious function. The rest groups were not obvious enough to provide useful data; Compared Group C, Group D1 approved obvious function(P<0.05); Group D1 showed stronger decrease function on the carbon powder propulsion of rat intestine than Group D2 and Group D3(P<0.05).(2) The decocting quantity of Paeoniflorin :The decocting quantity of Paeoniflorin was 0.97mg/g in Cassia Twig group. 1.22mg/g Paeoniflorin can be decocted in the group which Cassia Twig and Radix Paeoniae Alba were combined in ratio 1:0.5. 1.25mg/g Paeoniflorin can be decocted in the group which Cassia Twig and Radix Paeoniae Alba were combined in ratio 1:1. When Cassia Twig and Radix Paeoniae Alba were combined in ratio 1:2, 1.07mg/g. Paeoniflorin were produced.Conclusion:(1)All the experimental groups show the effect on decreasing the carbon powder propulsion of rat intestine.(2)When Cassia Twig and Radix Paeoniae Alba were combined in ratio 1:0.5, there came the strongest effect.(3)When Cassia Twig and Radix Paeoniae Alba were combined together, the decocting quantity of Paeoniflorin was higher than just Cassia Twig.(4)Cassia Twig can promote the decocting quantity of Paeoniflorin.
