ObjectiveMany clinical reports told us that the treatment of gouty arthritis about the Xinhuang tablets is significant.Because it contains indomethacin. There are a lot of side effects about the Xinhuang tablets in the aspect of the gastrointestinal tract.Therefore,we use the the Chinese medicine components of Xinhuang tablets to do the research.The purpose is to provide the reference for gout attack prevention drugs and secondary development of Xinhuang tablets.Methods[1] 72 male ICR mice were divided into six groups, such as the normal group, the model group, the Chinese medicine components of Xinhuang tablets dose groups (23.75 mg/kg,47.5 mg/kg and 95 mg/kg),the Benzene Bromide Malone group (20mg/kg). Mice administered orally once a day until 14 days. Groups of mice by intraperitoneal injection of oxygen oxazine acid potassium 300 mg/kg except the normal group at the final 1h after lavage. Detection indicators were the contents of uric acid in mice serum.[2] 108 male ICR mice were divided into nine groups such as the normal group, the model group, the Chinese medicine components of Xinhuang tablets dose groups (23.75 mg/kg,47.5 mg/kg and 95 mg/kg), the Allopurinol group (20 mg/kg). Mice administered orally once a day until 14 days. Groups of mice by intraperitoneal injection of hypoxanthine 1 g/kg except the blank group at the final 1h after lavage. Detection indicators were as follows: ①the content of uric acid in mice serum.②the content of xanthine oxidase in mice serum and liver. ③the expression of xanthine oxidase mRNA detected by RT-PCR in the liver of mice.[3] 90 male Wistar rats were divided into nine groups such as the normal group, the model group, the Chinese medicine components of Xinhuang tablets dose groups(23.75 mg/kg,47.5 mg/kg and 95 mg/kg), the Colchicine group (1.5 mg/kg). Rats administered orally once a day until 7 days. All the groups were acute gouty arthritis model except the normal group at the final lh after lavage. Detection indicators were as follows:① the general situation of rats.② the content of nitric oxide in serum of rats.③ the content of IL-1, IL-4, TNF-α in serum of rats and the content of MMP-3, IFN-γ, IL-4 In joint fluid of rats assayed by ELISA. Observe the joint and surrounding tissue, synovial membrane epithelium, inflammatory cells infiltration of each group rats by HE staining.Result[1] Compared with the normal group, the content of uric acid in the mice of the model control group increased significantly (P<0.01).The model was established successfully. Compared with model group, the Chinese medicine components of Xinhuang tablets can significantly reduce the blood uric acid levels in mice (P<0.05 or P<0.01)[2] Compared with the normal group, the content of uric acid and xanthine oxidase in the mice of the model control group increased significantly (P<0.01). The model was established successfully. Compared with model group, the Chinese medicine components of Xinhuang tablets can significantly reduce the blood uric acid levels and the content of xanthine oxidase in mice (P<0.05 or P<0.01). The RT-PCR result tell us that the Chinese medicine components of Xinhuang tablets can obviously cut the expression of xanthine oxidase in mice liver (P<0.01)[3] Compared with the normal group, the content of nitric oxide, IL-1, TNF-a, MMP-3, IFN-y in the rats of the model control group increased significantly (P<0.01). The content of IL-4 in the rats of the model control group reduced significantly. The model was established successfully. Compared with model group, the Chinese medicine components of Xinhuang tablets can significantly reduce the content of nitric oxide, IL-1, TNF-α, MMP-3, IFN-γ and increase the content of IL-4 in the rats (P<0.05 or P<0.01).Pathological histology results showed that model group rats synovial massive hyperemia, swelling, necrosis and inflammatory cells infiltration. But the Chinese medicine components of Xinhuang tablets can obviously reduce synovial tissue damage.ConclusionThe Chinese medicine components of Xinhuang tablets has a good curative effect for treatment of high uric acid fact. Its mechanism can lower the serum uric acid levels of mice and inhibit the xanthine oxidase activity. The Chinese medicine components of Xinhuang tablets is able to improve the mechanism of gouty arthritis of rats possibly by inhibiting the inflammatory markers and reducing the blood level of uric acid and nitric oxide.The results tell us that the Chinese medicine components of Xinhuang tablets can be a gout attack prevention drug and to provide the reference for clinical studies.
