Objectives: To investigate the causal relationship of DEHP with reproductive effects on male animals,and by different doses of DEHP effects on reproduction in male rats, explore DEHP affect male reproductive system in rats mechanisms and dose-effect relationship. DEHP provide reference for the study of the impact of human reproduction.Methods: On the basis of fed corn oil containing different concentrations of DEHP in female rats, The 28 pregnant rats were randomly divided into four groups. Gavage fed from the first day of pregnancy until the end of lactation. The rats were divided low-dose group(50mg / kg), the middle dose group(250mg /kg), the high-dose group(750mg / kg), Daily by gavage feeding program. Postpartum first day, according to the anogenital distance chosen for the study of male rats like, to all the animals were sacrificed on day 70 postpartum.Observe the growth and development of the various groups of pups, pups PND4 day, PND21 day survival, Prepulial Separation time and weight, testis and epididymis organ coefficient(organ weight/weight*100%), sperm count,viability sperm and motility sperm, serum testosterone and estradiol levels, testis and epididymis morphological changes and in the testis of P450 aromatase gene expression,comparative analysis.Results: By comparing each group’s growth and development of offspring rats, PND4 day and PND21 day survival,rats preputial separation time and rats preputial separation weight,newborn rats testis and newborn rats testis epididymis organ coefficient(organ weight / body weight * 100%), sperm count, viability sperm and motility sperm, serum testosterone and estradiol levels and testis P450 aromatase gene expression content difference:1.Birth weight offspring of high-dose group, medium and high dose groups anogenital distance significant difference compared with the control group; 2.High dose group PND21 offspring survival,testicular coefficient, medium and high dose group offspring body weight significant difference compared withthe control group; 3.PND35 PPS time when the high-dose group compared with the control group, the difference is obvious; 4.PND70 high dose group offspring body weight,testis and epididymis coefficien, epididymis sperm density,Middle and high dose groups of viability sperm, motility sperm, P450 Arom gene expression content compared to the control group significantly different; 5.DEHP in serum testosterone levels in male rats influential, It showed some dose-effect relationship between the T concentration and DEHP; 6.Test pups PND4 survival, PPS weight, E2 levels compared with the control group without significant difference.Conclusion: 1. DEHP impact on growth and development and reproductive capacity in male rats, DEHP can delay and inhibit the growth of rat reproductive capacity in rats.2. Different doses of DEHP could inhibit serum testosterone levels in rats, while a dose-response relationship.
