Enterovirus71 (enterovirus71, EV71) is a non-enveloped single-stranded positive-strand RNA viruses belonging to Picornaviridae genus. It is one of the main causing infant HFMD (hand-foot and mouth disease; HFMD) pathogens. Clinically EV71 can cause hand, foot and mouth diseases in infants and induced severe neurological diseases. Its pathogenesis remains unclear at now. EV71 virus replication studies have reported the synthesis of nucleic acids and proteins in cytoplasm, but the exact mechanism of its replication in the cell cytoplasm and sub cellular sites of replication, but so far, there is no relevant studies and reports. Through this study, we prove EV71 replication in the cell cytoplasm, and the RNA duplex observed in the intermediate (double-stranded RNA, dsRNA) formed during viral replication is present in the particle morphology of the cell cytoplasm structure. Further, we found in the cytoplasm, during viral replication, dsRNA formed with the lipid droplets (lipid droplet, LD) and lipid droplets surrounded protein (perilipin A) may be co- positioned with the time after infection and that of positioning the particles will gradually increase. Application of lipid droplets formed blockers triacsin C (acyl -CoA synthetase specificity inhibitor) and lipid droplets inhibitor C75 (fatty acid synthase, FAS), TOFA (adipose differentiation-related protein specific inhibitors) after 4h before infection we observed after pretreatment of the cytoplasm of infected cells, the localization of the particles significantly reduce the number of particles at different points in time, and no longer with the prolongation of the time of infection. Meanwhile, pretreatment cells infected cell supernatant virus titers were significantly reduced compared with the control. The first study found that EV71 virus replication may be associated with lipid metabolism, EV71-infected cells, the RNA replication may be mainly carried out in the lipid droplets in the cytoplasm. Relevance can be effectively suppressed after the formation of a specific blocker of lipid droplets, thus inhibiting the proliferation of EV71 replication.
