Objective:To explore the effects of 1,25?OH?2D3 on lipid droplets growth and cytokines in 3T3-L1 adipocytes of hypertrophy model.Methods:Cocktail method was used to induce differentiation on 3T3-L1 cell line.8 days later,cell modeled by 100,300,600,900?M PA for 24 h,cell without model set for control.The best concentration of modeling was screened by MTT and TG content.The model cells were intervened by 1,10 and 100 nM 1,25?OH?2D3 respectively,for 24 h,control group and model group were set for control.Then,cells were detected TG content and stained by oil red.The diameter and quantity of lipid droplet?LD?in cells were statistic by software Image-pro plus 6.0.mRNA relative expression level related to LD and TG metabolism?CIDE-a,Fsp27,PLIN-1,PPAR-?,PPAR-?,VDR,ACSL3,AGPAT1,ATGL,DGAT1,DGAT2,GPAT3,GPAT4,HSL,MGAT,PAP and UCP-1?were detected by RT-qPCR.The contents of TNF-?,IL-6,MCP-1,leptin,and adiponectin in cell supernatant were detected according to the kit.Result:1.According to the results of MTT and TG,300?M PA was selected as the optimum concentration.2.Compared with model group 10 and 100 nM 1,25?OH?2D3decreased the average diameter,increased the quantity of LD,up-regulated the mRNA expression level of PPAR-?and PLIN-1?P<0.05?,down-regulated the mRNA expression level of CIDE-a and Fsp27,significantly?P<0.05?.However,1 nM 1,25?OH?2D3 did not alter the morphology of LDs and TG content,as well as the expression level of related genes mRNA.There was no significant difference in the expression of PPAR-?and VDR mRNA between the 1,25?OH?2D3 intervention groups and the model group?P>0.05?.3.The mRNA expression levels of ACSL3,AGPAT1,DGAT2,GPAT3 and GPAT4 in the 10and 100 nM groups were significantly lower than those in the model group?P<0.05?;the mRNA expression levels of ATGL,DGAT1,HSL,MGAT,PAP and UCP-1 were significantly increased in the 100 nM group?P<0.05?.4.The levels of IL-6,MCP-1,TNF-?,leptin and adiponectin in the 1,25?OH?2D3 intervention groups were significantly lower than those in the model group?P<0.05?.Conclusion:The 10 and 100 nM1,25?OH?2D3 can inhibit LDs fusion,promote LDs decomposition,reduce LDs volume,inhibit lipogenesis through the PPAR-?signaling pathway,and may indirectly reduce the expression of IL-6,TNF-?and MCP-1.
