Objective: through the study on compensatory expression of aquaporin(Aquaporin, AQP) 3 in rat colon after Two-thirds resection and the effects of Traditional Chinese medicine, validate that if the remaining colon will compensate the moisture absorption by increasing the expression of the protein, as well as if this phenomenon is promoted by Pulsatilla Decoction. Therefore, we can provide a basis and direction of treatment for the symptoms of diarrhea patients in clinical colon resection.Methods: Select 27 adult male SD rats, randomly divided into control group(A), the experimental group(group B), Chinese intervention in the experimental group(group C), n = 9.Group A is sham-operation group; while group B and group C were done the extensive colon resection and end to end anastomosis surgery under general anesthesia. All three groups of rats were bred in single cages routinely and completed anti-infective therapy for three days; where group C was infused Pulsatilla soup for one week. Respectively, after 1,2, and 4 weeks, drawn the colon specimen from the same part of each rat which was selected randomly from those three groups; and immediately frozen in liquid nitrogen,-80℃ cryopreservation.Then detected the expression of colon AQP3 based on immunohistochemistry and real-time quantitative polymerase chain type reaction(Real-Time RT-PCR) methods and requirements, finally made a comparison of corresponding results by the appropriate statistical methods.Results: Compare with group A, the expression of Aquaporin 3 in group B and group C was increased on the first week, the second week and the forth week respectively(P<0.05); where group C were increased more significantly during the first week(P<0.05); but on the following second week and forth week, in comparison of group B and group C(P>0.05).Conclusion: The expression of Aquaporin 3 in rat colon has been increased, and at the meantime, Chinese medicine plays an role in promoting the compensation of Aquaporin 3.
