OBJECTIVES To analyze the effect of ODAM on the proliferation of thecolorectal cancer cell by transient transfection, stable transfection and proteinpurification methods, in order to further understand the mechanism of thepathogenesis and development of colorectal cancer.METHODS (1)MeasurethemRNAexpressionofODAMinthecolorectalcancercell lines HCT8、COLO205、HCT116and SW480;(2) Perform the proliferation assayafter transfecting transiently ODAM into HCT8;(3) Perform the proliferation assayafter transfecting stably ODAM into HCT8, including interference and overexpression;(4) Perform the proliferation assay after adding ODAM recombinant protein withdifferent concentrations into HCT8.RESULTS (1) The mRNA expression of ODAM in HCT8was higher than theother3lines;(2) After transfecting transiently ODAM into HCT8successfully, theproliferation of the colorectal cancer cell was increased;(3) After interfering stablyODAM successfully, the proliferation of the colorectal cancer cell was increased; afterover-expressing stably ODAM successfully, the proliferation of the colorectal cancercell was inhibited;(4) The proliferation assay showed that the proliferation of thecolorectal cancer cell was decreased with the concentrations of ODAM recombinantprotein growing. |
