LncRNA ENST00000430471 Promotes The Progress Of Colorectal Cancer By Regulating The Expression Of YBX-1

ObjectiveLong noncoding RNA(lncRNA)plays a crucial part in the progression of colorectal cancer.The high expression of ENST00000430471 and Y box binding protein 1(YBX-1)has been reported to be involved in colorectal cancer(CRC)due to its cancer-promoting ability.However,the detailed molecular mechanism of ENST00000430471 and YBX-1 in CRC is still currently unknown,which needs further study.In this study,we investigated the interaction between ENST00000430471 and YBX-1 and its role in the development of colorectal cancer.Methods1?Specific small inhibiting(si)RNAs were used to knockdown the expression of ENST00000430471.2?CCK-8 and Xenograft model were implemented to detect the effect of ENST00000430471 on tumor proliferation.To investigate the role of ENST00000430471 in migration and invasion,wound healing assays and Matrigel assays were performed,respectively.3?RNA pull-down assay with biotinylated ENST00000430471 was performed to identify the proteins interacting with ENST00000430471,followed by analysis using MS.4?To validate the role of the critical protein in ENST00000430471-mediated biological function in the progression of CRC,rescue experiments were performed.Results1?Small interfering RNA-si0471#2 was found to have the optimal interference efficiency in HCT116 and SW620.2?Knockdown of ENST00000430471 inhibited cell proliferation both in vivo and in vitro.Meanwhile depletion of ENST00000430471 suppressed cell migration and invasion in CRC.3?RNA pull down assay and mass spectrometry revealed that ENST00000430471 interacts with YBX-14?Knockdown of ENST00000430471 expression could decrease YBX-1 expression.5?YBX-1 overexpression could reverse the tumor suppressor effects produced by the knockdown of ENST00000430471 expression.ConclusionsENST00000430471 promotes the development of colorectal cancer by regulating the expression of YBX-1.
