Methods169male SD rats were randomly divided into sham-operation group, model group, Ginaton group and shi da la zhi wan group,the ischemic model was created by right middle cerebral artery occlusion(MCAO),ischemia reperfusion after2h, according to the time points after12h,24h,72h,7d group, a total of13groups, each group of13. Postoperative the sham-operation group and model group were given Physiological saline,the Ginaton group was given Ginaton,the shi da la zhi wan group wasgiven shi da la zhi wan.The neurological behavior scores of operation at12h、24h、72h and7d,4%formamint-fixed and decapitation brain. using Zea Longa and GarciaJ H observation nerve function defect degree of recovery;pathological changes were observed microscopically in HE stained sections;Apoptosis was detected by TUNEL method;The changes of expression of PI3-K、AKT、Bcl-2and Bax were detected by immunohistochemistry method.Results①C ompared with those of sham-operation group, the model group were significantly lower at each time point on neurological deficit scale scores(P<0.01);Compared with those of model group,the shi da la zhi wan group and Ginaton group wereevidently superior at each time point on neurological deficit scale scores(P<0.01);Compared with those of Ginaton group,hui the shi da la zhi wan group were significantly higher at72h,7d time point on neurological deficit scale scores(P<0.01).②The apoptoticcells of model group on different time points significantly increase compared with that of sham-operation group(P<0.01);the apoptotic cells of Ginaton group on different ti me points significantly decreased compared with that of model group(P<0.01);the apoptotic cells of Shi Da La Zhi Wan group on different time points significantly decreased compared with that of Ginaton group(P<0.01).③Compared with those of sham-operation group,the model group was significantly higher on PI3-K and AKT expression(P<0.01);Compared with those of model group,the each drug group was significantlyhigher on PI3-K and AKT expression(P<0.01);Compared with those of Ginaton group,shi da la zhi wan group was significantly higher on PI3-K and AKT expression, significantly at24h,72h,7d(P<0.01).④Compared with those of sham-operation group,the model group was significantly higher on Bcl-2and Bax expression(P<0.01);Compared with those of model group,the each drug group was significantly higher on Bcl-2expression(P<0.01),the each drug group was significantly decreased on Bax expression(P<0.01);Compared with those of Ginaton group,shi da la zhi wan group was significantly higher on Bcl-2expression(P<0.01),shi da la zhi wan group was significantly decreased on Bax expression(P<0.01).Conclusion shi da la zhi wan can improve the neural functional recovery in rats,TInhibition of neural cell apoptosis,the PI3-K and Akt was increased,activate the PI3-K/AKT signaling pathway may be associated to To reduce nerve cell apoptosis and protected Brain cells because of shi da la zhi wan.
