Objective: To research the regulated effect of electro-acupuncture at Dazhui point and Neiguan point on the apoptosis of the focal cerebral ischemia reperfusion model rats' brain tissue nerve cell and the expression of nerve growth factor(NGF)and glial cell-line derived neurotrophic factor(GDNF).Methods: Eighty SD rats were divided into 10 groups randomly. The groups included normal group, sham operation-24h group, sham operation-48h group, sham operation-72h group, model-24h group, model-48h group, model-72h group, EA-24h group, EA-48h group and EA-72h group. The focal cerebral ischemia reperfusion model rats were made by the method of improved plug in arteria cerebri media with thread. To observe the change of the rats' nerve function both after made models and treated by EA and before made models and treated by EA. TUNEL method was used to observe the apoptosis of the rats' hippocamp nerve cell and the effect of EA in each group. The effect of EA on the expressions of NGF and GDNF in brain tissue were observed by immunohistochemical method(SABC method). Results:1. Rats in model groups and EA groups show neurologic impairment physical sign in different degree after made model. Nerve function score show that there are significant differences between EA groups and model groups in the same phase(P<0.05,P<0.01). There are significant differences between EA-72h group and EA-24 group(P<0.01).These show EA can improve cerebral ischemia rats' nerve function and exist additive effect.2. Detection of TUNEL staining method: Compared with normal group and sham operation group, there are significant differences in model groups' number of dyeing positive cells(P<0.01). In EA-48h group, there are more TUNEL in ischemical hippocamp zone and the number reaches to the top. In EA-72h group, the number of dyeing positive cells reduces. There are significant differences between EA group and model group at the same phase(P<0.05). This show EA can inhibit apoptosis of brain nerve cell after focal cerebral ischemia.3. The result of image analysis of ischemia side brain tissue NGF immunological reaction positive cells shows the number of ischemical hippocamp zone NGF immunity positive expression cells in model groups increases obviously. Compared with sham operation groups, there are significant differences(P<0.01). This show the expression of NGF in brain tissue rise after ischemia. 48h group reaches to the top. There are...
