Objective To observe the expression level of neuropeptide Ghrelin in blood and braintissue in amygdala-kind led rat after seizure and to explore its causes and significance.Methods Methods Sixty-four healthy male SD rats were randomly divided intoamygdala-kindled group (n=48), sham-operated group (n=8) and blank control group (n=8).No treatment was performed in the blank control group. Only implantation of electrodes wasperformed in the sham-operated group. Implantation of electrodes and eletrophotolumi-nescence were performed in the amygdala-kindled group to induce amygdala-kindled seizuremodles. Rats in the amygdala-kindled group was divided into6sub-groups (n=8) at differenttimes after kindling (3,6,12,24,48, and72h). Enzyme linked immunosorbent assay,immunohistochemistry staining and real-time quantitative polymerase chain reaction wereemployed to detect the altered Ghrelin in blood and brain tissue of amygdala-kindled rat afterseizure.SPSS17.0software was used for statistical comparisons of data,and data expressed asthe means±SEM..Statistical comparisons of the means from different time points wereconducted using one-way ANOVA and Least-significant difference.Results (1) Behavior of Amygdala-Kinded rats and seizures situation: All amygdala-Kinded rats were reached V grade seizures, initially as shaking whiskernodding, eyes closed,salivation, hemifacial spasm, rhythmic chewing, followed by wet dog-shakes, forelimbelevation, convuls-ions immediately falls, seizures severely, there may be a sudden jumpand rapidly rotation. Seizures time of was about15seconds to several minutes.Afterdischarge threshold of Amygd-ala-kindled group is(112.7±27.43)μA, Speed of kindedis(7.12±2.13)d. (2)Concentration of octanoyl-greline in serum in Amygdala-kinded rats: No singnificantdifference on expression of acylated ghrelin was noted between the sham-operated groupand blank control group (P>0.05). As compared with sham-operated group,serum octanoyl-ghreline levels decreased significantly in difference time groups(P<0.05). There werestatistically significant(P<0.05) in difference time groups, between3h group and6h,group,between6h group and12h group, between24h group and48h group, between48h and72hgroup. There was no significant difference between12h group and24h group.(3)The expression of hypothalamus protein after seizures in Amygdala-kinded rats: Nosignificant difference on protein expressions of octanoyl-greline was noted between thesham-operated group and blank control group(P>0.05). As compared with sham-operatedgroup,the expression of hypothalamus protein decreased sig-nificant in difference timegroups(P<0.05). There were statistically sigificant in difference time groups(P<0.05),between3h group and6h group,between6h group and12h group, between12h group and24hgroup, between24h group and48h group, between48h group and72h group.(4)The expression of hypothalamus Ghrelin mRNAafter seizures in Amygdalakindedrats: No significant difference on mRNA expressions of hypothalamus Greline was notedbetween the sham-operated group and blank control group(P>0.05). As compared withsham-operated group, the expression of hypothlamus Ghreline m-RNA decreased signifycantin difference time groups(P<0.05).There were statistcally significant in difference timegroups(P<0.05), between3h group and6h group,between6h group and12h group,between12h group and24h group, between24h group and48h group. There was no significantdifference between48h group and72h group (p>0.05).Conclusion The expression level of acylated Ghrelin in serum could reflect level ofacylated Ghrelin in hypothalamus. It could be one of Clinical indicators on diagnose andtherapy assessment of epilepsy.
