Objective:To investigate the activation of ACE2-Ang-(1-7)-MAS receptors influe-nce on myocardial remodeling and the expressions of Ang II after myocardial infa-rction.Methods:SD rat were divided into sham-operated control group(n=8),myocard-ial infarction group(MI,n=8),losartan small-dose group (MI+10mg/kg·d for4weeks,n=8),losartan high-dose group(MI+20mg/kg·d for4weeks,n=8).Then established myo-cardial infarction model,detected the changes of electrocardiogram in Intraoperative. After4weeks,observed the pathological changes of myocardial by HE staining,Eac-h group of the myocardial tissue Ang-(1-7) content were detected by Elisa.the exp-ression of ACE2, MAS receptors and Ang-(1-7) in myocardium were detected by i mmumohistochemical staining, RT-PCR and Western Blot.Results:Myocyte swelling, deformation and fracture increase significantly in MI group compare with those in control group and alleviate in losartan small-dose group and further more decrease in losartan high-dose group.The expressions of Ang Ⅱ significantly upregulate in M I group than in control group,and downregulate in losartan small-dose group and fu rther more decrease in losartan high-dose group.The expressions of ACE2, Ang-(1-7) and MAS are slightly higher in MI group than in control group and increase in1osartan small-dose group and further more elevate in losartan group.Conclusion:My ocardial remodelling formed by increasing expression of Ang Ⅱ in MI group,itcan be inhibited by the increase expression of ACE2%Ang-(1-7) and MAS receptor.
