Objective:To discuss the role of PRP on the proliferation and osteogenetic activity of BMSCs in vitro culture and hormone intervention, and provide experimental basis for PRP technology in the application of the hormone treatment in ONFH patients.Methods:Rabbit model of glucocorticoid-induced ONFH was established through the experiment,and BMSCs intervened by hormone was obtained.The bone marrow cells of the rabbit models which were built successfully were cultivated in vitro and extended respectively.The rabbit BMSCs which had been extended and purified were divided into A and B groups.,then amplificated and cultivated in96well culture plate.A group(experimental group) was joined the mixed culture containing1.5%volume fraction PRP,and B group was joined the common culture.The sample drawn on a regular basis was tested by MTT,measured ALP activity and counted calcified nodules.Results:MTT:In4days A group was0.685±0.023,B group was0.399±0.021,the two groups had statistical difference (P<0.05),A group entered into the rapid proliferation period in advance.In6days A group was1.017±0.063,B group was0.704±0.019,the two groups had statistical difference (P<0.05),A group entered into the plateau proliferation period while B group was still in the proliferation stage.Until the8th day, the proliferation of B group was gradually approaching the platform period. ALP activity:In2,4,6,8day of the training,ALP activity in A group were significantly higher than that in B group.Calcium nodule count:A group was146.4±29.4/Fluoroscope vision,B group was82.6±24.5/Fluoroscope vision, the two groups had statistical difference (P<0.05).Conclusion:In vitro culture condition,the PRP can promote the proliferation and differentiantion in the direction of osteogenesis of BMSCs intervened by hormone.
