ObjectiveTo study the function and mechanism of the NgR-Rhoa-Rock1signal pathways which exists in the retinal ganglion cells in diabetes (diabetes mellitus, DM) rats.MethodsGetting healthy SD rats, we use a single intraperitoneal injection of50mg/kg wight1%preparation of streptozotocin DM model, and then randomly devided into3groups A, B and C, each set of10. In addition to take10healthy SD rats were compared.and only as a group D. Final respectively for all of4groups of rats were bilaterally eyeball intravitreal injection with NGR siRNA virus10μl, virus diluted10μl, virus-negative contrl solution10μl, virus diluted10μl, and been normal fed. After12weeks, slices for HE staining immunofluorescence staining、tunnel-stainig and to detect the expression of NgR-Rhoa-Rock1by Western blot analysis.ResultsWestern detection:compared with normal control group, the expression of NgR and Rhoa in DM group and siRNA-blank group increased significantly(p<0.01), while siRNA group no significant change(p>0.05); Compared with DM group and siRNA blank group, siRNA group protein expression significantly lowered.HE staining:compared with normal control group, three sets of model rats to some extent ganglion cells arranged disorder, irregular shape, spacing is not consistent;Compared with DM group and siRNA blank group, siRNA group of ganglion cells in the order, shape size and improved to a certain extent. Immunofluorescence staining:proteins distributes on the retinal ganglion cell layer massively. TUNEL staining:compared with normal control group, three sets of model rats were courteous, apoptosis.Compared with DM group and siRNA blank group, siRNA group of apoptotic cells is less, have improved significantly.ConclusionsThe expression of NgR and Rhoa, inhibition of NgR Rhoa-Rock can improve diabetic retinal ganglion cell apoptosis.
