Objective:To investigate the association between GRM8gene SNP and schizophrenia in the Uygur Chinese population. Methods:Subjects were reruited from Xinjiang Uygur Chinese population.The case-control study was used for the research. There were723schizophrenia patients and561non patients. The general demographic information and clinical data were collected, and then the database was established by Excel soft.The genomic DNA samples were extracted.2SNPs located in the GRM8gene were selected for genotyping by Taqman assays and capillary sequencing technology. The statistical analysis of allelic and genotypic frequency between2groups were carried out by the online software SHEsis,and Meta-analysis was carried out to identify the studies by software Review Manager5.2. Results:Both of the SNPs were not found deviation from Hardy-Weinberg equilibrium (P>0.05). For rs2299472, a statistically significant difference was observed between the2groups (P=0.015), and after Bonferroni’s correction, the difference still existed(P=0.03).The frequency of CC genotype was higher in patients (P=0.008),and the frequency of AC genotype was lower in patients (P=0.008). But there was not difference between the2groups in allelic frequency for rs2299472. The frequencies of rs2237748genotype and allele between the2groups were no significant difference (P>0.05). The meta-analysis also showed CC and AC genotypes were associated with schizophrenia(CC/AC+AA:P=0.0006;AC/AA+CC:P=0.002).Conclusion:①SNP19(rs2299472) located in GRM8gene is associated with schizophrenia in the Uygur Chinese population.②GRM8gene may play an important role in the pathogenesis of schizophrenia.③The DNA specimen database and the clinical database of Uygur Chinese population with schizophrenia was built up initially.And it provides reliable, high-quality resources for further genetic studies.
