Objective:To explore association between microsatellite D6S274 and D6S1960 polymorphism and schizophrenia by studying distributions of allele frequencies of the two loci between patients with schizophrenia and random population in Jiangxi Province.Methods:DNA samples were collected from patients with schizophrenia (n=174) in the Jiangxi Provincial Mental Hospital and healthy physical examinees(n=174) as controls matched with gender, age and race from the Jiangxi Provincial Chinese medicine hospital.Alleles and genotypes of the microsatellite D6S1960 and D6S274 were detected by the polymerase chain reaction-capillary electrophoresis,then they were analyzed by SPSS 16.0 statistic software,that is, first we calculated the allele gene and genotype frequency of the microsatellite D6S1960 and D6S274 for Hardy-Weinberg equilibrium testing goodness of fit to the control group,then compared the differences of the allele gene frequency of the microsatellite D6S1960 and D6S274 and analyzed odds ratios(OR) calculated by Chi-square test or fisher exact test(significance levelα=0.05, two-sided test).Results:3.1 Of the control group,observed values and expectated values of D6S1960 and D6S274 alleles were test by Hardy-Weinberg equilibrium testing goodness of fit,each loci were consistent with Hardy-Weinberg equilibrium (P>0.05).3.2 The frequency of the allele 273bp of D6S1960 in schizophrenia group was higher than that in control group (21.55%vsll.78%,χ2=11.96,P<0.01).The frequencies of other alleles did not differ between the schizophrenia and controls (P>0.05).3.3 The frequency of the allele 172 bp of D6S274 in schizophrenia group was higher than that in control group (11.78%vs4.60%,χ2=11.94,P<0.01).The frequencies of other alleles did not differ between the schizophrenia and controls (P>0.05).Conclusion:The microsatellite D6S1960 and D6S274 of the chromosome 6p are association with schizophrenia,it shows that gene associated with susceptibility to schizophrenia may exist in the microsatellite regions of D6S1960 and D6S274.
