Objective:To study the pharmacognosy of Stamens Mesua ferrea Linn.and provide basis for the further research of Stamens Mesua ferrea Linn.and thoughts in Multi-Components Library usa as a source of reference, to establish the Uygur Medicine Stamens Mesua ferrea Linn. Multi-component library (MCL), discusses its component separation and characterization. Methods:1. the pharmacognostical character of Uyghur medicine Stamens Mesua ferrea Linn.were studing by character identification, microscopic identification, physical and chemical identification and TLC identification methods.2. In vitro platelet aggregation test (turbidity) to evaluate the effect of different extraction of Stamens Mesua ferrea Linn platelet aggregation, and explore its biological characterization;3. Prepared the alcohol extracted components of Stamens Mesua ferrea Linn, and the Silca gel colum compenent were eluated with different concentration of petroleum ether acetone gradients.Using ODS reversed-phase column fraction was obtained with different concentration of Methanol water gradients and the Crystals were obtained by recrystallization method. The crystals were identified by Application of NMR technique.The n-butanol fractions were prepared under optimum chromatographic condition.The concentration of mobile phase, flow rate, sample volume and the detection wavelength was the main index, select the optimum chromatographic conditions.the extraction of components were analysised by using the HPLC-PDA technology, analysis the main absorption peaks in the chromatogram. The component analysis of HPLC graph and its antiplatelet activity combining results, to observe the relationship between component chemical characterization and biological characterization. Results:1. Stamens Mesua ferrea powder dispersed in pollen grains, clusters of calcium oxalate, threaded conduit, non glandular hairs, number is more, visible paracytic type, glandular hairs, square or polygonal parenchyma cell. Initially identified may contain steroidal, fats and oils, phenols, flavonoids, anthraquinones, cardiac glycosides, coumarins in Uygur Medicine Stamens Mesua ferrea Linn.Preliminary determined the oleanolic acid was identified by TLC.2. In vitro platelet aggregation experiment shows that, the petroleum ether, ethyl acetate, n-butanol and water fractions were different degree of inhibition of platelet aggregation, the n-butanol fraction is more significant, n-butanol fraction of10μ g/mL showed significantly inhibited by (P<0.01).3. There were19faractions prepared by silca gel column, in which the chemical structure of the obtained crystals was identificated by silica gel TLC and NMR data and found that the compound las chain sterols, compound2for shikimic acid, compound3for β-sitosterol, compound4was5,7-dihydrxy-6-(2-methylbutanoyl)-4-phenyl-2H-chromen-2-one, Compound5was5,7-dihydroxy-6--4-phenyl-2H-chromen-2-one,Compound6was1-cyclohexene-1-carboxylic acid-5-hydroxy-3,4-isopropylidene-two oxygen,5compounds were obtained from the total extract of Stamens Mesua ferrea Linn n-butanol fractions, using by liquid chromatography system, through the NMR identified for ethylshikimate compound7, compound8was isovitexin.4. The Hyperoside, ellagic acid and quercetin were detected for the first time from he ethylacetate extract using by HPLC-PDA technology. Conclusion:Preliminary Pharmacognostical studies study on the Uygur Medicine Stamens Mesua ferrea linn., to provide the reference for the identification and the further development and utilization. Through chemical constituents studies and foud of8compounds, in which Compound5was a new compound, named5,7-dihydroxy-6-(3-methylbutanoyl)-4-phenyl-2H-chromen-2-one. Using in vitro antiplatelet aggregation experiments to evaluate that n-butanol fraction activity more significantly, and it was closely related the ellagic acid and Hyperoside components etc. The research supplied data information for construction of Multi-Components Library of Uyghur Medicine Stamens Mesua ferrea Linn.
