The plant Swertia was a traditional medicine which called “Zang-Yin-Chen” in folk, forthe treatment of acute icteric hepatitis, cholecystitis, pneumonia, influenza, tonsillitis,osteomyelitis and other diseases. Medicinal value of the plant Swertia was given moreattention by researchers both at home and abroad, following with research of natural medicineand development of new drugs. In order to clarify the chemical foundation of this herb, thedissertation described the preparation, isolation and characterization of bioactive principlesfrom Swertia yunnanensis. The main research results of the article were as follows:HPLC analytical methods of Swertia were initially set up, by researching SwertiaYunnanensis, Swertia davidii, Swertia diluta, Swertia franchetiana. The results showed: Theanalysis methods were performed on a SinoChrom ODS-BP C18(4.6mm×250mm,5m);The mobile phase was acetonitrile-water(1000mLwater:0.2mLH3PO4:1mL NaOH(0.5mol/L)), mobile phase grads:010min(10%25%),1035min(25%),3545min (30%),4560min(30%65%); The flow rate was1mL/min; The detection wavelength was at254nm. And chemical components were separated effectively. Four kinds of swertia werecompared in similarity in this method, the result showed that the similarity of Swertiayunnanensis and Swertia davidii close to1.0, Swertia diluta and Swertia franchetiana close toeach other, but pre-post two groups were different in components. At the same time, thecontent of mangiferin of four kinds of Swertia were determined by using of HPLC, the orderof the content were Swertia diluta>Swertia franchetiana>Swertia yunnanensis> Swertiadavidii. If traditional chinese medicine could be replaced each other, a theoretical basis wasprovided by the research of similarity and determination of content of some component.Methanol extracts of Swertia yunnanensis were analyzed by using of HPLC, and varioustypes of compounds were classified, the separation of the objectives were identified at last.Methanol extracts of the herb were divided into five parts by Cyclohexane, Dichloromethane,Ethyl acetate and Methanol. Each polar portion was chemical characterized by HPLC-PDA.As a result, seven components were separated and prepared from the parts by using ofdifferent column chromatography techniques, such as normal phase and reversed-phase preparative chromatography. The structures were identified by MS and1H NMR and13CNMR, they were β-sitostero, Oleanolic acid,1,7-dihydroxy-3-methoxyxanthone,1,3,5,8-tetra-hydroxyxanthone, bellidifolin, swertianolin and mangiferin.
