Part Ⅰ Effect of the effective parts of Salvia chinensis of the biological behaviors of vascular endothelial cellsObjective:To investigate the effects of the effective parts of Salvia chinensis of the biological behaviors of vascular endothelial cells Methods:Human umbilical vein endothelial cells (HUVECs) were cultured primarily in vitro; The MTT assay was used to observe the effect of the effctive parts of Salvia chinensis on HUVECs and human lung carcinoma A549cell; Transwell cabin test and tube formation assay were used to observe the effect of the effctive parts of Salvia chinensis on the migration and tube formation ability of HUVECs. Results:3.125μg/ml~25μg/ml chloroform and ethyl acetate extract from Salvia chinensis role in HUVEC48h, the proliferation inhibition rate was17.36%to53.08%, from9.53%to31.36%。Acting on sensitive cell lines of human lung cancer A549proliferation inhibition rate of7.10%to31.18%,2.24%to21.69%, both compared to the significant differences in cytotoxicity (p=0.001)。3.125~25μg/ml chloroform and ethyl acetate extract from Salvia chinensis role the HUVECs12h migration inhibition rates were5.39%and57.38%,1.92%to37.37%;3.75μg/ml to25μg/ml trichloromethane extraction part from Salvia chinensis for24h HUVEC tube formation number reduced, and the lumen is not complete, the same concentration Iwami wear ethyl acetate extract weak inhibition; Conclusion:The trichloromethane extraction part from Salvia chinensis inhibit endothelial cell proliferation, migration and tube formation was significantly better than the ethyl acetate extract for the Salvia chinensis to play a major site of action of anti-angiogenic. PART Ⅱ Effect of the effective parts of Salvia chinensis of CAM angiogenesis inhibition in vivo experimental studyObjective:observation of tumor cell-induced angiogenesis in CAM phenomenon, explore the effective parts of Salvia chinensis inhibition of angiogenesis in CAM. Method:in vivo chicken chorioallantoic membrane (CAM) model, the observation of the inhibitory effect of the drugs on the CAM neovascularization; tumor cell inoculation in the exposed CAM, observation of tumor cell-induced angiogenesis, and observe the drug on tumor cell-induced angiogenesis inhibition. Results:3.125~25μg/ml chloroform and ethyl acetate extract from Salvia chinensis of CAM neovascularization rejection rate of10.24%to59.45%,-0.26%and12,2%, The trichloromethane extraction parts from Salvia chinensis inhibits angiogenesis stronger role; tumor cells induced angiogenesis in CAM capabilities, The trichloromethane extraction parts of200μg/ml of tumor cell-induced angiogenesis in CAM significantly inhibited, the ethyl acetate extract for the Salvia chinensis was not observed to significantly inhibit. Conclusion:The trichloromethane extraction parts from Salvia chinensis play a major site of action of anti-angiogenic.
