Objective:To observe Cathepsin S and NF-κB p65in the expression ofabdominal aortic aneurysm, and explore their roles in the rupture ofabdominal aortic aneurysm.Methods:Surgical specimens of the abdominal aortic aneurysm walltissue from80cases were collected, including36cases in AAA group,15cases in RAAA(<5cm) group,29cases in RAAA (Ruptured abdominalaortic aneurysm,≥5cm) group. Immunohistochemical and Western blotwere applied to detect the expression of Cathepsin S and NF-κB.Sperman method was applied for statistical analysis.Result:Immunohistochemistry showed that NF-κB p65expression wasfound in the cytoplasm and nucleus while Cathepsin S was mainlyconcentrated in the cytoplasm. NF-κB p65in RAAA (<5cm) positive ratewas significantly higher than the group AAA group, with statisticallysignificant differences (P <0.05), the RAAA (≥5cm) group, the positiverate higher than the AAA group, with very significant difference (P <0.01), Cathepsin S in RAAA (<5cm) positive rate was significantly higher thanthe group AAA group, with statistically significant differences (P <0.05),while its RAAA (≥5cm) group, the positive rate higher than the AAAgroup, with statistically significant differences (P <0.05),The expression ofboth Cathepsin S and NF-κB p65in abdominal aortic aneurysm tissues wascorrelated with hypertension and coronary artery disease (P <0.05). NF-κBp65expression was also associated with smoking (P <0.05).. Western blotshowed that NF-κB p65and Cathepsin S in RAAA (<5cm) group washigher than AAA group, with statistically significant differences (P <0.01),the group expressed in RAAA (≥5cm) above AAA group, with statisticallysignificant differences (P <0.01). Correlation analysis showed that theexpression of NF-κB p65and Cathepsin S was positively correlated (P<0.05).Conclusion: Cathepsin S and NF-κB p65were expressed inabdominal aortic aneurysm, and their expression lower in unrupturedabdominal aortic aneurysm,highly in ruptured abdominal aortic aneurysmtissue,was no correlated with the diameter in ruptured abdominalaortic aneurysm,while the expression of NF-κB p65and Cathepsin S waspositively correlated, suggesting that they might play an important role inthe rupture of abdominal aortic aneurysms.
