Objective:To explore the effect of thalidomide on the atherosclerotic plaque inWistar rat.Methods:Randomly selected10from40male Wistar rats as normal control group (groupA), given ordinary feed; The rest of the30rats performed operation of rataortic intima balloon injury, postoperative rats were randomly dividedinto three groups, respectively for atherosclerosis group(group B), low concentrationof thalidomide group(group C) and high concentration of thalidomide group(group D).all given a high-fat feed, and at the beginning of feeding by intraperitoneal injectionof giving vitamin D360IU/kg, injection once every1month later, constructing Asanimal model. Group C and group D postoperative lavage administration began toSally degrees amine, dose of10mg/kg respectively. D and30mg/kg. D.All of theanimals were executed at12weeks. And then collecting blood samples from thehearts to detect the lipid profiles,for example, TG TC, LDL-C, HDL-C and so on,Moreover, the level of TNF-αand IL-6were measured by enzyme linkedimmunosorbent assay (ELISA) in serum. Furthermore, taking the whole aorta, usingHE staining to observe the degree of plaque lesion, testing the thickness of aorticintima and tunica media, and the expression of TNF-α and IL–6detected byimmunohistochemical staining in atherosclerotic plaque.Results:1. Compared with group A, the blood-lipoid indexes of TG,TC, LDL-Cin groupB, C, D were significantly increased (P <0.05), but the blood-lipoid indexes ofTG, TC, LDL-C, HDL-C in group B, C, D were no significant difference (P>0.05).2. Compared with group B, the lipid content,the number of foam cells and inflammatory cells of plaque in group C were significantly reduced,and thethickness of aortic intima lower(P <0.05), tunica media thicken(P <0.05). Thedegree of pathological changes in group D was less than group C,it was showed thata small amount of foam cells and inflammatory cells infiltration,the thickness ofintima thin(P <0.05),the thickness of tunica media increase(P <0.05)。3. The levels of TNF-αand IL–6in serum of group D were lower thangroup B and group C(P <0.05), and group C were lower than group B (P <0.05).4. Immunohistochemical staining showed that the the expression levels ofTNF-αand IL-6in group D were lower than group B and group C (P <0.05),and group C were lower than group B(P <0.05).Conclusions:Thalidomide inhibit the atherosclerotic lesion could be throughthe anti-inflammatory effect, it is not related to lipid metabolism.
