Objective To explore the effects of growth and function of differenttiter of mycobacteriophage Chy3on human airway epithelial cell16HBEand preliminary assess the security of phage Chy3on human airwayepithelial cell16HBE, so as to further study animal experiments and lay afoundation about phage Chy3becoming a new biological agents withindependent intellectual property rights in the treatment of tuberculosis.Methods Inverted microscope was used to observed cellularmorphology,while cell viability and apoptosis rate were verified by CCK-8assay and flow cytometry. Additionally, the secretion levels of IL-6/IL-8in cell culture supernatant were detected by ELISA, RT-PCR and westernblot were performed to detect MUC5AC mRNA and protein expression.Results We found that the difference of the cellular morphology、cellviability、apoptosis rate、secretion levels of IL-6/IL-8、MUC5AC mRNAand protein relative expression of the cells treated with different titer of Chy3were no statistical significance compared to normal group (P>0.05).Conclusion These results indicate that there are no significant effectof growth and function of mycobacteriophage Chy3regardless of high andlow titer on human airway epithelial cell16HBE in vitro.Chy3in terms ofhuman airway epithelial cells has better security.
