Objective: To explore the related signal transduction mechanism inacid-induced expression of MUC5AC mRNA by airway epithelial cells.Methods:16HBE cells were stimulated by Hydrogen chloride, andtreated with OGR1siRNA and an inhibitors to ERK(PD98059). Cells weredivided into8groups: a control group(pH7.4), an acidic stimulation group(pH6.4), acidic stimulation+OGR1siRNAgroup, pH7.4+OGR1siRNAgroup, acidic stimulation+control siRNA group, pH7.4+control siRNAgroup, acidic stimulation+PD98059group, pH7.4+PD98059group. Thetranscription level of mucin(MUC)5AC was evaluated with reversetranscription-polymerase chain reaction(RT-PCR).The MUC5AC secretedin supernatant was measured by enzyme-linked immunosorbentassay(ELISA). The expression of OGR1protein and The protein levels ofphosphorylation extracellular signal-regulated kinase (p-ERK) wasmeasured by Western blot.Results:The highest expression of MUC5AC mRNA and MUC5ACprotein in acidic stimulation group compared with the control group (p<0.05), The expression of OGR1protein and p-ERK were alsosignificantly different from those in control group, The expression ofMUC5AC mRNA and MUC5AC levels of phosphorylation ERK in acidicstimulation+OGR1siRNA group were also significantly decreased, Theexpression of MUC5AC mRNA and MUC5AC protein showed no differentbetween pH7.4+OGR1siRNA and the protein in acidic stimulation+OGR1siRNA group were lower than the acidic stimulation group(p<0.05), Theprotein control group(p>0.05), phosphorylation ERK also showed nosignificant change after transfection of OGR1siRNA, The secretion levelof MUC5AC and the expression of MUC5AC mRNA were much lower inacidic stimulation+PD98059group than the acidic stimulation group(p<0.05), while there were no significance between the a control groupand pH7.4+PD98059group in secretion level of MUC5AC and expressionof MUC5AC mRNA(p>0.05).Conclusion:OGR1/ERK transduction cascade may be upper signalregulators in acid-induced MUC5AC expression in16HBE cell.
