Objective:HCC(hepatocellular carcinoma) is one of the common tumors, miRNA is involved inthe regulation of oncogenes and tumor suppressor genes is one of the mechanisms of HCC.The experiment is to detect the expression of miR-331-3p in HCC cells and HCC tissues, toexplore the relationship between clinical pathological features of miR-331-3p and HCC andits clinical significance.Methods:1.The qRT-PCR technique was used to validate different expression ofmiR-331-3p in immortal liver cell line HL-7702and different invasion and metastasis ofHCC cell lines (the invasion ability: HepG2<MHCC97-H <HCCLM3).2.The qRT-PCR technique was used to detect expression and correlation analysis ofmiR-331-3p for5controlled liver tissues,30HCC tissues and adjacent tissues.3.To analyze the relationship between the expression of miR-331-3p and theclinicopathological characteristics of HCC.Results:1.In qRT-PCR analysis showed that, three human hepatocellular carcinoma cell lineswith different ability of invasion and metastasis (HepG2, MHCC97-H and HCCLM3) andone immortal liver cell line(HL-7702) had expressed the miR-331-3p. the average comparedwith single factor variance analysis; p<0.05for the difference was statistically significant.the expression of miR-331-3p in all kinds of cells was: HL-7702> HepG2> MHCC97-H>HCCLM3.2.In qRT-PCR analysis showed that, there was significant difference in expressionof miR-331-3p in hepatocellular carcinoma tissues, adjacent carcinoma tissues and controlledliver tissues (p<0.05); and compared with controlled liver tissues, the expression ofmiR-331-3p in HCC tissues is significantly decreased. 3. It showed expression of miR-331-3p in multiple tumor nodules, low differentiation,and venous invasion in the HCC tissues was significantly lower than that of single nodule(p=0.036), high-middle differentiation (p=0.035), no venous invasion(p=0.016) of HCCtissues. The expression of miR-331-3p of gender, age, tumor size, AFP value, with orwithout cirrhosis and presence of the envelope, there was no significant correlation(p>0.05).Conclusion:1. miR-331-3p in HCC cell lines (HepG2, MHCC97-H, HCCLM3) is down-regulate.2. The expression of miR-331-3p in HCC tissues is significantly decreased, which isrelated to multiple tumor nodules, low differentiation, and venous invasion.
