Objective:Observing the TNF-α changes after rats cerebral hemorrhage, exploring genisteinblocking EGFR-STAT3pathway effects on the expression of TNF-α levels in ratsafter cerebral hemorrhage.Methods:Take54healthy male SD rats, weighing250~280g, were randomly divided intothree groups: GEN group (18), cerebral hemorrhage (18), sham group (18). GEN group and intracerebral hemorrhage in stereotactic, caudate nucleus within its injectable collagenase+0.7uL0.14uIV heparin-induced intracerebral hemorrhage model obtained,resulting in sham group injected with saline dose. After the model was successfully manufactured in rats after intraperitoneal injection of GEN group75mg/kg daily genistein solution (dissolved in the GEN25ulDMSO solution), then the same amount of DMSO solution was intraperitoneally injected to replace GEN intracerebral hemorrhage group and sham group. In the postoperative1d,3d,7d three time points Zeal longa5neurological scoring method of rat nerve function score, score each time point in each group were sacrificed six rats per group, brain tissue slices, using immunohistochemicaldetection of the expression of EGFR, STAT3, and TNF-α in the rat brain hemorrhagesurrounding tissues, Western blot to detect the protein EGFR, STAT3, and TNF-α expression levelsResults:After intracerebral hemorrhage, intracerebral hemorrhage apparent neurological deficits performance and loss of appetite, weight loss, in the first three days about the worst performance, compared with its, GEN Zeal longa rats at3and7days neurol ogical scores5points lower, the difference was statistically significant (P <0.05), sham group neurological score unchanged. HE staining light microscope and found that intracerebral hemorrhage surrounding tissue proliferation of astrocytes activation GENgroup was significantly higher than sham group had no significant change. The results of immunohistochemistry and Western Blot results showed: Compared with the sham group, EGFR, STAT3and TNF-α expression in intracerebral hemorrhage group a-.nd the GEN group and are raised, the difference was significant (P <difference between expression and intracerebral hemorrhage0.05), one day after ICH, GEN group EGFR, STAT3, and TNF-α was not statistically significant (P>0.05); first three days of intracerebral hemorrhage EGFR, STAT3and TNF-expression of α peaked, then decreased gradually,7d still express, GEN group expression3d and7d rat brain tissue EGFR, STAT3, and TNF-α in lower than intracerebral hemorrhage group, the difference wasstatistically significant (P <0.05).Conclusion:1The EGFR,STAT3and TNF-α in rats brain upregulated after cerebralhemorrhage;2GEN may inhibit TNF-α expression in rat brain after intracerebral hemorrhageby inhibiting EGFR-STAT3pathway...
