Traditional Chinese Medicine (TCM) The Experimental Study Of Mice Allergic Contact Dermatitis

Objectives:Eczema is a kind of allergic inflammatory skin disease, belongs to the IV allergy. Its clinical manifestations and pathological characteristics are equal to contact dermatitis. So this study established allergic contact dermatitis model in Balb/C mice using the FITC1%(acetone: DBP1:1) and observed the pathological effect influcing by folium isatidis, golden cypress and gentian. To explore the mechanism of heat-clearing and damp-drying drug, heat-clearing and blood-cooling drugs in treating acute dermatitis eczema.Methods:1.35female Balb/C mice,three months old, were randomly divided into7groups, nameed blank control group, saline group, dexamethasone group, folium isatidis group, golden cypress group, gentian group folium isatidis and gentian mixed group.2. Experiments were made in0day with back shaving,1st day and2nd day with sensitization using FITC1%(acetone:DBP1:1) on mice back shaving,7th day with FITC0.5%acetone:DBP (1:1) on mice ear. All mice were treated in the4th day and were killed in the9th day to observe induced dermatitis ear thickness and ear pathological changes, etc.Results:1. Inspire1. Mice appeared different degree of swelling, or had mild erosion, or itching (the scratching performance) after stimulate each ear in.2. Compared with saline group, the ear thickness difference (S1) and the same mice ear thickness difference (S2) significantly decreased (P<0.01) in the other groups. Golden cypress group and folium isatidis and gentian group also have obvious difference (P<0.05).3. Compared with saline group, the edema in epidermis cells of mice significantly reduce in the other groups.4. The dermal edema decreased obviously in folium isatidis group, golden cypress group and gentian group. The dermal edema also has a certain reduce in folium isatidis and gentian group also has a certain reduce; capillaries expansion were reduced in the dermis in all group exceprt for golden cypress group 5.Compared with saline group, the degree of mononuclear cells infiltration decreased significantly in dexamethasone group, gentian group and folium isatidis group(P<0.01); gentian group, gentian group and folium isatidis group also had obvious changes (P<0.05); the degree of Polymorphism nuclear cells infiltration decreased significantly in gentian group and folium isatidis group (P<0.010.05).6.compared with dexamethasone group, there was no significant statistical difference in reducing mice ear skin in folium isatidis group (P>0.01); there was no significant difference of mononuclear cell count between folium isatidis group and dexamethasone group (P>0.01).7. Compared with dexamethasone group, there was no significant difference in inhibiting inflammation of mononuclear cell infiltrates in golden cypress group and gentian group.8. Compared with dexamethasone group, there was no significant difference in decreasing edema in inside and outside and dermal epidermal cells in folium isatidis group, golden cypress group and gentian group.Conclusion:1. Folium isatidis, golden cypress and gentian can effectively inhibit Ⅳ type of allergy of mice.2. Traditional Chinese medicine can achieve the same effects glucocorticoids in some aspects on inhibiting allergic contact dermatitis.
