The Distribution And Significance Of HoxA13from All Kinds Of Hypospadias

Objective:To investigate the role of the HoxA13gene from the HOX family in the development of hypospadias by detecting the transcription and expression of HoxA13in the prepuce and urethral plate of hypospadias patients.Methods:We collected the tissues from the prepuce and urethral plate of30hypospadias patients aged3.3～11.6years, the prepuce of10phimosis children aged3.1～10.4years and the urethra of10penile carcinoma patients aged48.1～75.6years with no urethral involvement, the latter20taken as controls. We divided the tissue samples into a distal, an intermedial, a proximal and a control group, and detected the expressions of HoxA13m R NA and protein in different groups byRT-PCR and immunohistochemistry.Results:RT-PCR:Transcriptase of HoxA13in the prepuce and urethral plate from Intermedial Group(urethral plate:0.684±0.228, Prepuce:0.848±0.338) and Proximal Group(urethral plate:0.464±0.164, Prepuce:0.497±0.218) reduced much significantly compared with Control Group (urethral plate:1.270±0.209, Prepuce:1.409±0.441)(P<0.05). Proximal Group also reduced significantly compared with distal Group(urethral plate:1.054±0.189, Prepuce:1.071±0.342)(P<0.05). IHC:Protein of HoxA13in the urethral plate and prepuce of Intermedial Group(urethral plate:5238±3065, Prepuce:5217±1993) and Proximal one(urethral plate:2238±2217, Prepuce:2095±1591) reduced significantly compared with Control Group (urethral plate:13420±2636, Prepuce:12050±4112)(P<0.05). Proximal Group also reduced much significantly compared with distal one(Prepuce:8223±3212, urethral plate:10450±2123)(P<0.05).Conclusions:There is high correlation between HoxA13gene in the prepuce and urethral plate of hypospadias and the kind of hypospadias. It may indicate that the extence of Hoxa13transcription and expression is concerned with the position of urethral meatus.
