Objective:Our research aims to study the distribution of matrix metalloproteinase3in adult rat retina and investigate the influence of MMP3for the survival of RGCs in adult rat retina following acute high intraocular pressureMethods and Materials:Adult SD rats were divided into five groups: HIOP group、HIOP+NNGH group、HIOP+3%DMSO group、control group and Sham-operated group.rats in HIOP group were injected normal saline to elevated the intraocular pressure to110mmHg and maintained for60min,; rats in HIOP+NNGH group and HIOP+3%DMSO group were intraperitoneal injected with NNGH (N-Isobutyl-N-(4-methoxyphenylsulfonyl)glycyl hydroxamic acid, A highly sensitive inhibitor of MMP3) and3%DMSO30min before the treatment of HIOP respectively, and repeated the injection every8h for one day. Rats in control group were not done with any process and rats in Sham-operated group were punctured by needle and maintained for the intraocular pressure to20mmHg. These rats were sacrificed at different time point (6h, Id,3d,7d). Western blotting, immunohistochemistry, immunofluorescence and double-labelling immunofluorescence were used to investigate the distribution of MMP3in adult rat retina and the influence of MMP3for the survival of RGCs in adult rat retina following acute high intraocular pressureResult:1:In adult rat retina, the positive products of MMP3were located in nerve fiber layer, ganglion cell layer, inner and outer plexiform layer. In the ganglion cell layer, the positive products of MMP3were double labeled with NeuN; at the edge of the inner nuclear layer, the positive products of MMP3were double labeled with Calbindin and Parvalbumin; in the inner and outer plexiform layer; In inner plexiform layer and inner nuclear layer,the positive products of MMP3were double labeled with PKC-α; the positive products of MMP3double labeled with Synaptophysin; while there was no MMP3positive products seen in the outer nuclear layer. In the nerve fiber layer and ganglion cell layer, MMP3was also double labeled with GFAP and GS; while in the inner plexiform layer, it was double labeled with CD11b.2:MMP3expression was elevated in the early stage of high intraocular pressure retinas and dropped after3d.3:The upregulation of MMP3expression was surpressed at1d and3d after acute HIOP by NNGH injection. With the injection of NNGH, number of the survived RGCs was significantly increased at3d after acute HIOP.Summary:In normal adult rat retina, Matrix metalloproteinase3mainly localized in most neurons and glial cells, while the up-regulation of MMP3expression may participate in the loss of RGCs after acute HIOP.
