| Abstract:obiectives:to study the expressionã€distribution and clinical pathological significance of GPER and Gankyrin in paired ectopic and eutopic endometrium of ovarian endometriosis and normal endometrium, so as to investigate the roles in the occurrence and development of ovarian endometriosis, and to provide a new theoretical basis for the pathogenesis and treatment of endometriosis.Method:We analysize the expression and distribution of GPER and Gankyrin protein and mRNA in paired ectopic and eutopic endometrium and normal endometrium through immunohistochemistry SPã€western blotting and Real-time PCR.Results:1. Real-time PCR results showed that both the relative expression level of GPER and Gankyrin mRNA in paired ectopic and eutopic endometrium of endometriosis and normal endometrium were0.0355ã€0.0050ã€0.0015and0.0521ã€0.0091ã€0.0052, the two mRNA among the three groups showed a great trend of decline (P<0.05). In ectopic endometrium, The relative expression of GPER and Gankyrin mRNA showed no statistical difference between â…¢-â…£ stage and â… -â…¡ stage (P>0.05).2. Western-blotting analysis results showed that both the relative expression level of GPER and Gankyrin proteins in paired ectopic and eutopic endometrium of ovarian endometriosis and normal endometrium were0.7968±0.0967ã€0.6926±0.1188ã€0.3922±0.0787and0.8144±0.1010ã€0.7293±0.1521ã€0.4135±0.1255, the two proteins among the three groups showed a great trend of decline (P<0.05). In ectopic endometrium, the relative expression level of GPER and Gankyrin proteins in â…¢-â…£ stage were higher than that in â… -â…¡ stage (P<0.05).3. Immunohistochemistry results showed GPER protein mainly localized on the membrane and cytoplasm of endometrial glandular epithelial cell, a little expressed in stromal cell. Gankyrin protein expressed mainly in the nucleus and cytoplasm of endometrial epithelial cells, some stromal cells were also positive. Both the relative expression level of GPER and Gankyrin proteins in paired ectopic and eutopic endometrium of ovarian endometriosis and normal endometrium were2.38±0.24ã€2.21±0.30ã€1.13±0.21and2.55±0.21ã€2.30±0.34ã€1.21±0.25. The two proteins among the three groups showed great difference (P<0.05). In eutopic endometrium of endometriosis and normal endometrium, the expression of GPER and Gankyrin were higher in proliferative phase than secretory phase (P<0.05). In ectopic endometrium, the relative expression of GPER and Gankyrin proteins in â…¢-â…£ stage were higher than that in â… -â…¡ stage (P<0.05).4. The Pearson correlation analysis showed that the protein expression of GPER was positively correlated with Gankyrin both in ectopic and eutopic endometrium of the ovarian endometriosis (P <0.05).There was no correlation between GPER and Gankyrin in normal endometrium(P>0.05).Conclusions:1. GPER and Gankyrin are highly expressed in paired ectopic and eutopic endometrium of ovarian endometriosis, both of which play important roles in the development of ovarian endometriosis.2. The expression of GPER and Gankyrin were relevant with different clinical stages of endometriosis, which indicates that they may be a promising indicator of evaluating the severity and progression of endometriosis.3. There was a positive correlation between the expression of GPER and Gankyrin proteins in paired ectopic and eutopic endometrium of ovarian endometriosis, which indicates that they may have a synergetic relationship to participate in the formation and development of endometriosis. |
