Objective:to study the expression of matrix metalloprotekinase-3(hereinafter to be referred to as MMP-3) and liver-intestine cadherin (also known as CDH17,hereinafter to be referred to as CDH17) in colorectal cancer, to study the relevance between them and the position of cancer tissue, degree of differentiation, TNM stage as well as patient’s age, gender. Meanwhile to explore the influence and clinical significance they play on the occurrence, development of colorectal cancer.Methods:Select50tissue samples, which are diagnosed as rectal adenocarcinoma in both preoperative and postoperative pathologic examination in Second Xiangya Hospital of Central South University and First People’s Hospital of Changde City from May to September2013, as the experimental group. Select30cases of normal tissue from5cm of cancer tissue of the experimental group as the control group. Use the method of tissue RNA extraction and fluorescence quantitative detection to study the expression level of MMP-3and CDH17in experimental group and control group. Perform the statistical analysis of experiment results using the SPSS.17software, to analyze if their expressions in cancer tissue and adjacent normal tissue are different by two independent samples T-test analysis; to analyze the relationship between their expressions and the position of cancer tissue, degree of differentiation, TNM stage as well as patient’s age, gender. Study the expression differences of MMP-3and CDH17in different tissue differentiation and TNM staging through the method of analysis of variance (ANOVA) or rank.Results:1the expression of MMP-3in colorectal cancer(1) The expression of MMP-3in colorectal cancer tissue is significantly higher than in normal tissue adjacent to carcinoma, and the difference is statistically significant (t=5.46, p<0.01);(2) The worse the colorectal cancer tissue differentiation is, the higher expression of MMP-3in colorectal cancer, and the difference is statistically significant (F=25.36, P<0.01);(3) The later the colorectal cancer tissue staging is, the higher expression of MMP-3in colorectal cancer, and the difference is statistically significant (F=31.06, P<0.01);(4) The expression of MMP-3in colorectal cancer tissue of low age groups is significantly lower than in high age groups, and the difference is statistically significant (F=7.09, t<0.01);(5) There is no obvious statistical correlation between the expression of MMP-3and the position of tumor (t=-0.209, P>0.05); (6) There is no obvious statistical correlation between the expression of MMP-3and patient’s gender (t=0.183, P>0.05)2the expression of CDH17in colorectal cancer(1)The expression of CDH17in colorectal cancer tissue is significantly lower than in normal tissue adjacent to carcinoma, and the difference is statistically significant (t=-10.07, P<0.01);(2) The worse the colorectal cancer tissue differentiation is, the lower expression of CDH17in colorectal cancer, and the difference is statistically significant (F=17.77, P<0.01);(3) The later the colorectal cancer tissue staging is, the lower expression of CDH17in colorectal cancer, and the difference is statistically significant (F=7.94, P<0.05);(4) The expression of CDH17in the colon cancer is lower than in rectal cancer tissue, and the difference is statistically significant (F=-3.25, P<0.05);(5) There is no obvious statistical correlation between the expression of CDH17and patient’s gender (t=1.70, P>0.05);(6) There is no obvious statistical correlation between the expression of CDH17and patient’s gender (F=1.20, P>0.05)3There is a positive correlation between MMP-3and CDH17, and they have significant statistical significance, since the correlation coefficient is0.376and P value is less0.01. Conclusions:No.1The high expression of MMP-3and/or low expression of CDH17is closely related to the development of colorectal cancer, and they play an important role in local infiltration, which helps the distant metastasis of tumor cells.No.2The high expression of MMP-3and low expression of CDH17act synergistically in local infiltration of colorectal cancer.No.3The abnormal expression of MMP-3and CDH17is expected to be an index which helps to assess patient’s condition, detect the recurrence and prognosis.
