The Expression OfTRAF1、4-1BB、Bcl-xL In The Tissue Of Gastic Mucosa Infected By H.Pylori And The Relationship With Virulence Genotypes Of H.Pylori

Objective:Our study group had found that the gene expression of TRAF1,4-1BB, and Bcl-xL increased in GES-1cell(Immortalized gastric mucosal cell) cultured with Hp (cagA+/vacAsl+/vacml+).To research the possible mechanism that promote the occurrence of gastric cancer under the condition of Hp (cagA+/vacAsl+/vacml+) infection, we detect the protein expression of TRAF1,4-1BB,and Bcl-xL in different pathologic stage of gastric cancer(gastritis, precancerosis, cancer) and the HP virulence genotype, and analyze the effect of TRAF1,4-1BB,and Bcl-xL in the process of transformation from gastritis to gastric cancer.Methods:1. Collecting patients infected Hp in different pathologic stage(chronic non-atrophic gastritis, Intestinal metaplasia with atypical hyperplasia, gastric cancer); using PCR and Western Blot) to detect the protein expression of TRAF1,4-1BB,and Bcl-xL and analyzing the mutual relation among three peroteins.2.Extracting the Hp DNA from the gastric mucosa tissue in different pathological stage, detecting the Hp virulence genotype and analyzing the Hp genotype distribution between up-regulation group of TRAF1,4-1BB,and Bcl-xL and non-upregulation group, and further analyzing the correlation of the protein expression of TRAF1,4-1BB,and Bcl-xL and Hp virulence genotype (cagA+/vacAsl+/vacml+).Results:1. Compared to chronic atrophic gastritis group, the protein expression of TRAF1,4-1BB,and Bcl-xL significantly increased in intestinal metaplasia with atypical hyperplasia group and gastric cancer group; mutual correlation among3proteins:the correlation coefficient between TRAF1and4-1BB is0.678; TRAF1and Bcl-xL is0.702,4-1BB and Bcl-xL is0.694.2. the genotype of HP in increased protein expression /vacAsl+/vacml+genotype distribution between increased group and non-increased group of different genes has statistical difference (P<0.05).Conclusions:1. TRAF1,4-1BB,and Bcl-xLproteins take part in the development process of gastric cancer which result from gastritis with Hp infection and might promote the evolution of gastric mucosal from gastritis to gastric cancer.2. cagA+/vacAsl+/vacml+Hp genotype infection might promote the protein up-regulation of TRAF1,4-1BB,and Bcl-xL, and further promote the process of gastric cancer which result from gastritis...
