ObjectiveRecently, TRAF have emerged as an important connection factor in signal transducers pathways. At present, six types TRAF family members have been discovered, their characteristics are nearly similar;and they can combine with many TNFR and IL - 1R family members, which actives NF - κB and JNK signal pathways. And they have some important function to regulate reproduction, survival, apoptosis and lead to sprouting of endothelial cell. TRAF2,3,6 expressed widely in normal tissue, but TRAF1,4,5 only expressed in some tissue. The study on TRAF expression and function in tumor is rare. This study investigated TRAF expression in breast cancer and the correlation between their expression and MVD.Methods(1) We examined TRAF expression in normal breast and breast cancer with immunohistochemistry.(2) We also examined TRAF expression with western blot in cultured high metastasis and low metastasis human breast cancer cells, respectively MDA -MB-231 and MCF-7.(3) Statistics with SPSS10. 0 software and appling with t - test and x~2 -test. P < 0. 05 means significance and P < 0. 01 means predominant significance.Results1. Examining TRAF expression with immunohistochemistry in breast cancer.(1) TRAF1 do not express in normal breast. Positive expression rate of TRAF1 in breast cancer increases to 28.57%. Compared with that in intraductal carcinoma, TRAF1 positive rate in invasive carcinoma increases notably(P<0. 01).(2) TRAF2 does not express in normal breast. Positive expression rate of TRAF2 in breast cancer increases to 37.14%. Compared with that in intraductal carcinoma, TRAF2 positive rate in invasive carcinoma increases slightly(P >0. 05).( 3 ) Positive expression rate of TRAF3 in breast tissue is low. TRAF3 positive rate in breast carcinoma increases notably (P <0.01) . Compared with that in intraductal carcinoma, TRAF3 positive rate in invasive carcinoma decreases notably(P<0.01).(4 ) TRAF4 expresses both in normal breast tissue and breast tumor tissue, and expression intensity between them has not outstanding difference ( P > 0. 05 ) . Compared with that in intraductal carcinoma, TRAF4 positive rate in invasive carcinoma increases slightly ( P > 0. 05 ) . TRAF4 nucleus positive rate is notable different between normal breast and breast cancer (P < 0. 01). Compared with that in intraductal carcinoma, TRAF4 positive rate in invasive carcinoma decreases notably ( P <0.05 ) .(5) Both TRAF5 and TRAF4 expression level in normal breast and cancer tissue are outstanding high, and expression degree between them is not differ-ence(P >0. 05). Compared with that in intraductal carcinoma, TRAF5 positive rate in invasive carcinoma increases slightly (P>0.05).(6) Positive expression rate of TRAF6 in breast cancer is low. TRAF6 positive rate in carcinoma increases notably (P <0. 05) . Compared with that in intraductal carcinoma, TRAF6 positive rate in invasive carcinoma increases notably (P<0.01).2. Examining TRAF expression in cultured human breast cancer cells with western blot.(1) Compared with that in MCF -7, TRAF1 A2A4^5^6 protein expression quantity in MDA -MB -231 enhances notably.(2) But TRAF3 protein expression quantity in MDA - MB -231 is less than that in MCF-7.3. Relations between TRAF and MVD.In invasive carcinoma, MVD of TRAF1 or TRAF5 is notable difference (P <0. 05) between their positive and negative expression. MVD of TRAF1 or TRAF5 positive expression is more concentrated than that of their negative expression.In intraductal carcinoma, MVD of TRAF3 is notable difference (P <0.05) between its positive and negative expression. MVD of TRAF3 positive expression is more concentrated than that of its negative expression.Conclusions1. TRAFs protein expression in breast cancer increase more than in normal breast tissue. Compared with that in normal breast, TRAF1 A2^3^6 positive rates in breast carcinoma increase notably.2. Compared with that in intraductal carcinoma, TRAFK6 positive rate in invasive carcinoma increases notably.3. TRAF4 nucleus positive expression rate in invasive carcinoma is lower than that in intraductal carcinoma.4. TRAF1^5 and MVD in invasive carcinoma is positive correlation.
