Objective:During this experiment,by testing the expression of miR-150,miR-155in plasma in patients with acute leukemia(AL), to analyse the expression of miR-150,miR-155in plasma in the different course, study its correlation with clinical features, explore its clinical significance in AL.Methods:Collecting the peripheral blood specimens of acute leukemia patients and healthy people as controls, extract total RNA of plasma, using the method of real-time fluorescent quantitative PCR to detect the relative expression levels of miR-150,miR-155,to compare the expression differences between different groups in patients with AL and healthy groups. Collecting the correlative clinical materials in patients with AL to analyze its correlation with clinical features. Build the ROC curves, through the area under the curve to analyze the sensitivity and specificity to diagnose AL through miR-150, miR-155in plasma.Results:1. Expression of miR-150in AL significantly decreased compared with healthy groups(p<0.05), expression of miR-155in AL significantly increased compared with healthy groups(p<0.05).2.Expression of miR-150between groups:expression of miR-150in patients with primary treatment AL and relapse AL significantly decreased compared with healthy and remission groups.(p<0.05). no difference was found between patients with primary treatment AL and relapse AL, as well as healthy and remission groups.2. Expression of miR-155between groups:expression of miR-155in patients with primary treatment AL and relapse patients significantly increased compared with healthy and remission groups.(p<.05),no difference was found between patients with primary treatment AL and relapse AL, as well as healthy and remission groups.3.In the primary treatment group,miR-150in patients with AML, ALL expression is significantly lower than normal control, but No difference was found between AML and ALL.4. In the primary treatment group,miR-155in patients with AML ALL expression is significantly higher than normal control, but No difference was found between AML and ALL.5.Expression of miR-150and miR-155in primary treatment AL has no correlation with age, sex, white blood cells、hemoglobin concentration,platelet count and blast count in the bone marrow。6.The ROC curves Show that the test of miR-150and miR-155and both for the diagnoses of AUC is89%,84%,93%respectively.Conclusion:1.compare with the normally control group, the expression of circulating miR-150in the acute leukemia decreased dramatically; while,the expression of mir-155increased significantly compared with normally control group, indicating that the abnormally expressed miRNA may take part in the development of acute leukemia.2.the expression of miR-150、miR-155in the remission was close to the control, may be used as indexes of evaluating the curative effect of chemotherapy; while the group showes the same changes with the initially treated group,may be used as the monitoring indexes of remission.3. the miR-150、miR-155have same variation tendency both in AML and ALL,have nothing to do with cell types4.this study indicates that the expression of miR-150、miR-155have nothing to do with age,sex, bone marrow cell number as well as peripheral blood indicators.5.the separated and united detection of circulating miR-150、 miR-155both have higher AUC value,and have higher diagnostic rats and united detection have higher sensitivity and specificity in the diagnose of acute leukemia.This article includes10tables,12figures and33references.
