Objective:Investigated the effect of miRNA125b in cisplatin inhibiting the proliferation of osteosarcoma MG-63cell.Methods:1. Detected the proliferation of osteosarcoma MG-63cell under the intervention of different concentrations’cisplatin by MTT assay.2. The osteosarcoma MG-63cells were divided into experimental group (with the intervention of cisplatin) and control group (without the intervention of cisplatin), then detected the expression of miRNA125b respectively.3. MiRNA125b were silenced and overexpressed in osteosarcoma MG-63cells,and divided the cells into four groups as miRNA125b mimic, miRNA125b inhibitor, miRNA125b NC and miRNA125b inhibitor NC, then detected the proliferation of osteosarcoma MG-63cell under the intervention of IC50concentration’cisplatin by MTT assay.Results:1. After conducting a number of MTT assay with the intervention of cisplatin, we could observe that with increasing the concentration of cisplatin in osteosarcoma MG-63cells, the inhibition rate increased. We got the IC50of osteosarcoma MG-63cell was26.73under the cisplatin inhibition by IC50calculation software.2. Osteosarcoma MG-63cells with IC50concentration’cisplatin intervention and no intervention incubated for48hours,then detected the expression of miRNA125b.We got the expression ratio of miRNA125b (experimental group/control group) was0.312in the intervention group. We could see that the expression of miRNA125b was down-regulated in MG-63cells with cisplatin intervention relatived to MG-63cells without cisplatin intervention.3. Measured the absorbance of each hole in the ELISA analyzer OD490nm place, calculated the inhibition rate,we could get that the inhibition rate of the miRNA125b inhibitor group was lower than the miRNA125b inhibitor NC group, the inhibition rate of miRNA125b mimic group was higher than the miRNA125b NC group, the difference was statistically significant (P<0.05).Conclusion:The upregulation of miRNA125b expression helped cisplatin to inhibit the proliferation of osteosarcoma cell, miRNA125b might be one of the mechanisms that cisplatin inhibited the proliferation of osteosarcoma cell.
