Objective:To study the difference of autophagy level between breast cancer and breast benign tumors, and the effect of chemotherapy for the autophagy level of breast cancer, and to analysis the possible relationship between autophagy and the effect of chemotherapy.Method:An immunohistochemistry was used to measure the expression of LC3and Beclinl before and after neoadjuvant chemotherapy for breast cancer tissues and breast fibroadenoma tissues. Analysis the differential expression of LC3and Beclinl between breast cancer tissues and breast fibroadenoma tissue, and analysis the expression change in breast cancer before and after chemotherapy. Then analysis the relationship by SPSS18.0software among the expression of LC3and Beclinl, clinical pathological features and chemotherapeutic effect.Results:Expression of LC3and Beclinl in breast fibroadenoma tissues was significant higher than breast cancer (P<0.01).After chemotherapy, Beclinl and LC3expression was significantly elevated (P<0.01). However, the expression of LC3and Beclinl did not have statistical significance in different patients with different clinical pathological features, such as age, menopausal status, clinical stages, estrogen receptor (ER), progesterone receptor (PR), axillary lumph nodes metastasis, etc. When40patients with neoadjuvant chemotherapy were divided into three group, such as partial response (PR,33patients), stable disease (SD,6patients) and progressive disease (PD,1patient), the LC3expression in PR group was significant lower than SD group before chemotherapy (P<0.05).Conclusions:(1) Compared with breast fibroadenoma tissues, down-expression of LC3and Beclinl in breast cancer indicates that autophagy level might be inhibited in breast cancer.(2) Up-regulation of LC3and Beclinl after neoadjuvant chemotherapy suggests that chemotherapy may improve the cell autophagy level.(3)Breast cancer with high level of autophagy is poor sensitivity to chemotherapy, which indicating that LC3may be a new tumor maker for predicating chemotherapy-sensitivity of breast cancer.
