Objective:To observe the effect of Abnormal Savda Munziq(ASMq) on learning and memory, blood biochemical index, immune function, morphological structure of heart, liver, brain, p16and β-galactosidas(SA-βGal) expression, expression of NF-κB p65mRNA in spleen on D-galactose induced aging model rat’s, in order to clarify the abnormal savda munziq part of anti aging mechanism action. Methods:take three month old SD rats, male(n=60), weighing180-220, used D-galactose induced subacute aging model and three different dosages (6.0,3.0,1.5g/kg·d-1) of ASMq is given to the model group, test to the ability of rets learning and memory, blood biochemical index.determination of rat serum IL-2、IL-6、TNF-α level.observe histological changes of heart, liver, brain.observe the effect of p16, β-galactosidas inheart, liver, brain and the expression of NF-KBp65mRNA in spleen was detected with RT-PCR.Results (1) compared with the aging model group, ASMq can improve significantly the ability of learning and memory in aging rets, the difference were statistically significant(p<0.05-0.01).(2) compared with the aging model group, there was no significant difference of ASMq high, middle and low dose group WBC, RBC, HGB, HCT, MCV, PLT, ALT, AST, TP, CRE, UA, ALB, GLU,(P>0.05).but there was have significant difference of ASMq high, middle and low dose group compared with the aging model group in TG, TC, HDL, LDL (P<0.05).(3) ASMq high-dose group compared with model group increased IL-2, decreased IL-6, TNF-α (P<0.01); ASMq medium-dose group compared with model group increased IL-2, decreased IL-6, TNF-a (P<0.01); ASMq low-dose group compared with model group increased IL-2, decreased IL-6(P<0.01), decreased TNF-a(P>0.05).(4) myocardial: compared with the aging model group, ASMq high, middle and low dose group myocardial cells plump, Myofilament neatly arranged, Appeared mildly vasodilation. brain:compared with the aging model group, ASMq high, middle dose group Structural integrity of cell morphology neatly arranged.There has been little vacuolar changes and karyopyknosis in the low dose group. liver:compared with the aging model group, ASMq high, middle and low dose group liver cells structural integrity. Have a small amount of lipid vacuoles and edema, but there was no karyopycnosis.(5) Compared with the normal control group, the expression of p16protein, SA-βGal in heart, brain, liver tissue of aging model group were obviously increased(P<0.05-0.01), Compared with the aging model group, the expression of β16protein, SA-βGal in heart, brain, liver tissue of (ASMq)high, middle and low dosage group were obviously decreased(P<0.05-0.01).(6) Compared with the normal control group, the expression of NF-KBp65mRNA in spleen of aging model group were obviously increased(P<0.01). Compared with the aging model group, the expression of NF-κBp65mRNA in spleen of (ASMq)high, middle dosage group were decreased(P<0.05-0.01). low dosage group decreased NF-KBp65mRNA, but the differences were not statistically significant (P>0.05). Conclusion:(1) the study conclude that ASMq have anti-aging effect.its possible mechanism may be by reducing expression of p16protein, SA-βGal and NF-κB p65mRNA.
