Objective: To observe the effect of Abnormal Savda Munziq(ASMq) on theserum,heart,brain and liver tissues free radical metabolism and endogenous metabolites inplasma on D-galactose induced aging model rat`s, on the free radical metabolism andMetabonomics level investigate the inherent anti-aging mechanism of ASMq. Methods:Aging rat model was induced by D-galactose and three different dosages (1.5,3.0,6.0g·kg-1) of ASMq is given to the model group to observe the concentrations of SOD,GSH-Px,T-AOC,NO and MDA on the serum,heart,brain and liver tissues. Plasma sampleshave been detected by biological NMR techniques, and the dates of NMR spectra havebeen analyzed by Pattern recognition analysis to find out the changed endogenousmetabolites in the rat`s plasma. Results:1. Compared to the normal control group, theconcentrations of SOD, GSH-Px and T-AOC were obviously decreased (p<0.01) and theconcentrations of MDA and NO were obviously increased (p<0.01) in serum,heart,brainand liver tissue of aging model group. Compared to the aging model group,theconcentrations of SOD, GSH-Px and T-AOC were obviously increased (P<0.05~0.01),and the concentrations of MDA and NO were obviously decreased inserum,heart,brain and liver tissue of three dosage ASMq groups(P<0.05~0.01),theconcentration of SOD in serum of low dosage ASMq group was no significantlydifference with aging model group;2. Compared to the normal control group,Theconcentrations of glycine, leucine, isoleucine, methionine, valine, lactic acid, inositol,creatine, carnitine, choline, acetic acid, pyruvic acid, α-glucose and β-glucose in plasmawas increased significantly(p<0.05) and the concentrations of Alanine, histidine, lipids(VLDL, LDL), an unsaturated lipid, acetone and glycoprotein in plasma decreasedsignificantly(p<0.05). Compared to the aging model group, The concentrations of glycine,isoleucine, methionine, valine, inositol, creatine, lactic acid, carnitine, choline, α-glucose, beta-glucose was decreased significantly(p<0.05),and the concentrations of Histidine,phenylalanine, alanine, lipids (VLDL, LDL), unsaturated lipids acetone,glycoprotein wasincreased significantly(p<0.05) in Plasma. Conclusion: Abnormal Savda Munziq canimprove antioxidant enzyme activity and prevent free radical oxidation, and reduce freeradical damages in aging rat model; abnormal savda munziq can adjust and improve thedisordered glucose metabolism, lipid metabolism, amino acid metabolism of aging ratmodel at some extent. regulation of D-galactose induced aging rats free radicalmetabolism and plasma metabolites is the main aspects of the anti-aging mechanism ofASMq.
